中文摘要 | 本研究自臺灣省西部地區土壤中篩選出一株黴菌,以CzA、C20S及 M40Y三種培養 基在 25、 37 及 50 ℃培養 10 天, 結果生長均相當良好, 因此將其界定為高溫菌 (thermophile),此菌且耐高滲透壓 (40% sucrose),菌落在 CzA 上培養 10 天時呈帶灰色 之橄欖綠,直徑為 5.5 ∼ 6.0 cm。顯微鏡檢查之結果,分生孢子頭緻密,呈柱狀,帶灰色 之橄欖綠,未見閉囊殼,分生孢子柄寬 5.7 μ m,長 64 ∼ 128 μ m,孢囊呈球形,直徑 14 ∼ 18 μ m,直立在分生孢子炳上,小梗為緊密的單層,分生孢子呈球形,多刺。 該菌 株之外觀形態與 Aspergillus fumigatus 相似,唯耐滲透性高。 因此暫定為 Aspergillus sp., 命名為 Aspergillus sp. NTU-FC-671。 Aspergillus sp. NTU-FC-671 可分泌高溫性 及非高溫性蛋白�t。 高溫性蛋白�t最適之反應溫度為 80 ℃,於 70 ℃加熱 1 小時之活性 保留為 88%, 最適作用 pH 為 5.0; 非高溫性蛋白�t之最適反應溫度為 50 ∼ 60 ℃,於 50 ℃加熱 1 小時之活性保留為 55%,最適作用 pH 為 8.0。 |
英文摘要 | A mold was isolated from soil by growing on CzA、C20S and M40Y media at 25 ℃、 37 ℃ and 50 ℃ for 10 days, respectively. Since it grew satisfactorily on the above media, it was defined as a thermophile. It also resisted high osmotic pressure. The colony on the CzA medium was grayish olive green at the 10th day with 5.5 ∼ 6.0 cm diameter. The conidial head was compact columnar with a grayisholive green color. The conidiophore was 5 ∼ 7 μ m wide and 64 ∼ 128 μ m long. The vescile showed a globose which had a diameter of 14 ∼ 18 μ m and stood on the conidiophore. The sterigmmta was a compact uniseriate. The conidia was ball-shaped and echi nulate. According to these morphologic characteristics, the mole was similar to Aspergillus fumigatus except for high osmotic pressure resistance. It was temporarily identified as an Aspergillus sp. and was named Aspergillus sp. NTU-FC-671. Aspergillus sp. NTU-FC-671 could produce thermostable and nonthermostable proteases. The optimum pH and temperature of the thermostable protease were 5.0 and 80 ℃, respectively. The thermostable protease retained 88% of its activity when it was heated at 70 ℃ for 1hr. The optimum pH and temperature of the nonthermostable protease were 8.0 and 50 ∼ 60 ℃, respectively. The nonthermostable protease retained 55% of its activity when it was heated at 50 ℃ for 1 hr. |
