中文摘要 | 本研究由產氣膨瓶醬油所分離四株酵母菌和包含這些菌屬之典型的菌株 Saccharomyces, Zygosaccharomyces 和 Candida 三個屬, 共 21 種,54 株菌,以兩對真 菌的普遍性引子, 藉聚合�t鏈鎖反應增殖核醣體 DNA 的內轉錄區 ITSI 和 ITSII。 在 Zygosaccharomyces 屬的 10 種菌中, 除了 Z. fermentati 和 Z. cidri ITSI 和 ITSII 片段長度無法區別外, 其餘相同種菌的 DNA 片段長度都相同,不同種菌的片段長度皆不同 , 因此可作為 Zygosaccharomyces 屬內菌種的鑑定方法; Candida 屬各菌種之 ITSI 和 ITSII 之片段長度非常接近, 無法作為屬內菌種之鑑定。 但 ITSI 經純化後以毛地黃素 (digoxigenin) 標識後為 DNA 探針,與本試驗之測試菌株雜交, 能快速而正確的鑑定產氣 膨瓶之酵母菌菌種。 |
英文摘要 | Four yeast species isolated from gas forming soy sauce and the other typical yeast strains including three genera-Saccharomyces, Zygosaccharomyces and Candida, with a total of 21 species and 54 strains, were investigated in this report. Two sets of universal primers were used to amplify ITSI and ITSII of ribosomal DNA by polymerase chain reaction (PCR). The same Zygosaccharomyces species produced identical fragment lengths, but different species except Z. ferentati and Z. cidri did not. In the experiments, eigth species of Candida produced fragments with similar sizes, so it is necessary of find another DNA probe to identify the species in Candida. According to the results, the PCR-product ITSI could be purified and labeled with digoxigenin as DNA probes and hybridized with typical yeast cultures to identify the yeast species correctly and rapidly. |
