中文摘要 | 實驗中將市售之五種常用中藥材(當歸.川芎.白朮.赤芍藥.燈心草)以適當之溶媒 抽取並濃縮成乾粉後, 分別與四種常用的賦形劑 (Microcrystalline Cellulose (Avicel PH 101), Corn starch, Dicalium Phosphate Dihydrate, Lactose) 以 1:1 (w/w) 的比例 混合。 所製得之樣品分別置於由二種, 相對濕度 (0%, 75%) 及三種溫度 (35 ℃, 45 ℃, 55 ℃ ) 所構成的六種環境中貯存,並定時取出以高效率液相層析儀 (HPLC) 測定其主成份 含量的變化情形。 各成份 HPLC 條件如下: 層析管柱: RP-C18(1) 當歸, 川芎 (cis N-butylidenephthalide):移動相:水: 氰甲烷:甲醇:冰醋酸 =500:120::450:20 ml 檢 出波長: UV 254nm 流速:1.5 ml/min(2) 白朮 (Atractylon):移動相:水:氰甲烷 =550 :450 檢出波長:UV 254nm 流速:1 ml/min(3) 赤芍藥 (Paeoniflorin): 移動相:水:氰 甲烷 =900:100 (含 0.03% 85% 磷酸 ) 檢出波長: UV 254nm 流速:1.2 ml/min 20 mins, 2.2 ml/mins(4) 燈心草:(Luteolin) 移動相:甲醇:水:冰醋酸 =450:550:1 檢出波長: UV 254nm 流速:1.5 ml/min 對於一些具有芳香油的中藥 (ex. 當歸. 川芎 )。尤需特別注 意保存的條件吾人相信適當的保存條件比適當的賦形劑更重要。 赤芍藥和白朮在 DSC 上均 顯示指標成分與 DCP. lactose 間存在有交互作用其安定性資料亦顯示其較不安定,其他賦 形劑則差異不大。 燈心草在 DSC 和安定性資料上顯示似乎很單純,和賦形劑之間似乎無明 顯之交互作用,濕度之影響較不明顯,看來是以 starch 對安定性最有幫助。本實驗中並利 用示差掃描熱分析儀預測藥品主成份與賦形劑間的配伍性,並與含量分析結果對照。結果顯 示兩者確有關係存在。由安定性資料顯示,在高濕度的環境下,有效成分的衰減十分明顯, 這個結果提醒我們一件事情就是 -- 中藥應保存在乾燥處所,潮濕會導至藥品效價變差。 |
英文摘要 | Five Chinese herbs, Angelicae Radix, Cnidii Rhizoma (Ligustici Rhizoma) , Junci Medulla, Atractylodis Rhizoma, and Paeoniae Radix were extracted with suitable solvents, then the extracts were freeze dried The subsequent dry powders were separately mixed with four commonly used excipients, microcrystalline cellulose, corn starch, dicalcium phosphate dihydrate and lactose in 1:1 (w/w) ratio, respectively. The subsequent mixtures were stored at various conditions of different relative humidities and temperatures for investigating the effects of various excipients as well as the storage conditions on the stabilities of the active ingredients. The active ingredient of each herb in the mixture was analysed at fixed time intervals using an HPLC method. The compatibilities between the active ingredients and the four respective excipients were also studied usinig a differential scanning calorimetry (DSC) method. The results showed generally that corn starch was the most suitable excipient for these herbal extracts. A higher relative humidity had a more significant effect on the declined profiles of the major ingredients of these five extracts during storage. |
