中文摘要 | 本試驗嘗試探討並建立臺灣肖楠(Calocedrus formosana(Florin)Florin)之 組織培養之微體繁殖體系。初步結果:臺灣肖楠表面消毒試驗之結果,種子可達 90 %之無 污染率, 未成熟胚則可達零污染,4 ∼ 6 個月實生苗莖段可控制在 92.5 %無污染率。 2 ∼ 3 年生實生苗莖段控制污染僅能達到 57.5 %之無污染率, 具培植體於消毒後多褐化死 亡。在器官形成方面:成熟胚培植於 WPM、MS、B �N三種基礎培養基對無菌苗之誘導效果最 好。 未成熟胚培養於添加 600 mg/l casein hydrolysate、500 mg/l yeast extract、0.5 mg/l BA 及 1 mg/l NAA 之 WPN 培養基中,可誘導出多芽體。無菌苗帶節莖段在添加 0.05 mg/l NAA 與 0.5 mg/l BA 之 WPM 培養基中可誘得最多腋芽之發生。4 ∼ 6 個月實生苗針 葉帶節莖段在含有 0.05 mg/l NAA 及 1 mg/BA 處理可誘導腋芽之發生, 鱗狀葉培養於 1 mg/l BA 與 0.05 mg/l NAA 之 WPM 培養基中可誘導形成腋芽,但芽體叢生狀並玻璃質化, 而在含有 0.01 mg/l BA 與 0.05 mg/l TDZ 之處理可獲得 60 %之不定芽誘導率。 癒合組 織在含有 1 mg/l NAA 及 0.1 mg/l BA 之 MS 培養基誘導效果最佳。 |
英文摘要 | This report describes the development of in vitro culture method for Calocedrus formosana (Florin) Florin. The primary results are summarized as follow. Mature embryos, immature embryos, needle-like leaves and scale-like leaves from 4-6 month-old seedlings, and shoot tips from lateral branchesof 2-3-year-old trees were used as explants. In surface sterilization, immature embryos were decontaminated completely. Ninety percent of the seeds were sterilized. Explants from 4 to 6-month-old seedlings were 92.5 % sterile. Although 57.5 % of the explants derived from 2 to 3-year-old trees were decontaminated, none of the explants survived in the subsequent culture. The MS, WPM and B �N were the three best media for raising sterile plants. In vitro multiple shoots were successfully induced from immature embryos cultured on the WPN medium containing 600 mg/l casien hydrolysate, 500 mg/l yeast extract, 0.5 mg/l BA and 1 mg/l NAA. Bud formation was optimal on WPN medium supplemented with 0.05 mg/l NAA and 0.5 mg/l BA when nodal segments from sterile seedligs were cultured. Needle-like leaves with nodal segments excised from 4 to 6-month-old seedlings could induced bud formation when culturing on WPN medium containing 0.05 mg/l NAA and 1 mg/l BA. Although bud formation could also obtained from scale-like leaves cultured on WPN medium containing 1 mg/l BA and 0.05 mg/l NAA, buds were clumped and vitrified. However, bud formation was increased to 60 % if scale-like leaves were cultured on WPN medium supplemented with 0.01 mg/l BA and 0.05 mg/l TDZ. Callus induction was optimal on MS medium containing 1 mg/l NAA and 0.1 mg/l BA. |
