中文摘要 | 馬匹的繁殖管理和遺傳型研究是引種的重要工作項目,本研究應用臺灣省畜產試驗所畜牧場的馬匹為試驗材料,進行血液採樣,測定其葡萄糖磷酸基異構?(GPI)和六磷酸鹽葡萄糖去氫?(PGD)的遺傳型以及紅血球膜上的葡萄糖六磷酸鹽基去氫?(G6PD)和血清乳酸去氫?(LDH)的活性。分別有來自美國的小型馬家族八匹、迷你馬家族七匹和來自澳洲的迷你馬家族六匹受測,合計21匹的GPI遺傳型均為BB型,而PGD遺傳型在小型馬家族和迷你馬家族有AB型和AA型個體。酵素活性測定上未有家族的顯著性差異存在,G6PD值介於105~185mU/e9R,LDH值介於510~943 U.L。馬匹的年齡和酵素活性未有明顯的關係存在,不過在此較同父異母的澳洲迷你馬時,G6PD和血紅素之比值(G6PD/Hb)有隨年齡增加略呈下降的趨勢。澳洲迷你馬家族之G6PD/Hb為10.1±2.3mU/g,而美國迷你馬和小型馬家族則為11.9±3.0mU/g和11.4±2.6mU/g,暗示馬匹的遺傳生理值不受地區來源、品種和年齡影響。本研究建立的鑑別技術可供其他馬匹族群應用。 |
英文摘要 | Genotyping and reproductive management of horses after introduced are essential to the propagation of the new breeds. The present study was conducted by collecting the blood samples of horses kept at the Animal Farm of Taiwan Livestock Research Institute. The genotypes of glucose phosphate isomerase (GPI) and 6-phosphogluconate dehydrogenase (PGD) were then identified with electrophoresis. The enzymatic activity of glucose-6-phosphate dehydrogenase (G6PD) on the red blood cell membrane and serum lactic dehydrogenase (LDH) were measured. A total of 21 horses were grouped into three by breed characteristics and countries of origin, which included the American pony family (N=8), the American miniature hourse faily (N=7) and the Australian miniature horse family (N=6). The GPI genotypes of all 21 horses were BB. The PGD genotypes were PGD-AA and PGD-AB genotypes in pony and miniature horses. The enzymatic activity of G6PD or LDH among three families were not significantly different. The activity of G6PD varied with the measurements of 105~185 mU/e9R and so to 510~943 U/L for LDH. Both G6PD and LDH enzymatic activity were not significantly correlated with the age of horse. In the paternal half-sib miniature horse family imported from Australia, the ratio of G6PD/hemoglobin decreased slightly with the increase of age in miniature horses. Ratios of G6PD/Hb were 10.1±2.3mU/g, 11.9±3.0mU/g and 11.4±2.6mU/g in the Australia miniature horse family, the American miniature horse family and pony family, respectively. Results indicated that the physiological measurements of horses were not affected by their countries of origin, breeds and ages. Hence, the genotyping techniques and measurement of enzymatic activity could be applied for other horse populations. |
