中文摘要 | 本報告以遺傳工程技術、基因重組及脂轉形(lipofection)的方法,製造出假性狂 犬病毒台南株醣白蛋白gE(以前稱之gI)基因缺損之突變株。初步以墨點雜交法(dot blot hybridization)篩選出幾個可能的病毒株,其中一株,命名為R23,更進一步以南方雜合試 驗(Southern blotting)及西方點墨法(western blotting)進行確認,結果顯示此R23株之醣 白蛋白gE基因已被刪除,完全無法製造出gE的蛋白質。以動物實驗測試之初期結果,亦可 顯示此R23株具有免疫功能,應可預防豬隻之假性犬病毒強毒株之感染,以期開發為疫苗之 用。 |
英文摘要 | The glycoprotein E(previously named gI) deletion mutant of pseudorabies virus strain TNL was generated by employing recombinant DNA technique. Initial screening was done by dot blot hybridization and several possible mutant clones were obtained. One mutant clone, designated as R23, was further examined carefully by Southern and western blottings, and the results showed that the clone R23 was deleted in gE gene and did not express glycoprotein gE at all. Preliminary animal test suggested that R23 was immunogenic and conferred immunity on pigs against virulent pseudorabies virus. |
