中文摘要 | 本研究主要目的在探討維生素B6是否可減緩胰島素治療的糖尿病血管病變。以Streptozotocin(STZ)誘導糖尿病大白鼠,分別施以胰島素、維生素B6或胰島素與維生素B6同時治療。以腺磷雙磷酸(ADP,10μM)或凝血(Thrombin,0.05U/mL當致凝物,在血小板凝集儀下測定血小板凝集程度。以放射性花生四烯酸(14C-Arachidonic Acid, �釮-AA)為前驅物,利用放射性薄層層析法(Radiochromatography Scanning)及自動放射照像術(Autoradio-graphy)分析血小板生成前列凝素(�釮-ThromboxaneB��,�釮-TxB��)的情形,並用放射線閃爍計數器(Scintillation Counter)測量�釮-TxB�砥A生成量。實驗結果發現,胰島素與維生素B6同時治療的實驗組,血糖值低於胰島素或維生素B6單獨治療的兩組。同時血小板凝集程度及血小板生成前列凝素程度亦有顯著降低。以上結果皆顯示,維生素Bfi可減緩胰島素治療糖尿病大白鼠之病情與血管併發症的誘因,對血糖控制、防止血小板凝集、減少前列凝素生成皆比胰島素單獨治療的效果好。 |
英文摘要 | The purpose of this study is to investigate whether vitamin B6 alleviates thevascular complications of insulin-treated streptozotocin(STZ)-induced diabetes inrats. Diabetic animals were treated with or without vitamin B6 and/or insulin.Platelet aggregation induced by ADP (10 μ M) or thrombin (0.05 U/mL) wasmeasured in platelet rich plasma of normal and diabetic animals. �釮-Thromboxane B�� (�釮-TxBg) production of platelets, using �愔釮-Arachidonic Acid(�釮-AA) asa precursor, was assayed by means of scanning radiochromatography and autoradiography. �釮-TxBz was quantified by scintillation counter. The results showedthat vitamin B6 in conjuction with insulin treatment resulted in lower bloodglucose than either vitamin B6 or insulin treatment alone. Similarly, plateletaggregation and TxB�� production in diabetics with vitamin B6 and insulin treatment were significantly decreased. These data indicated that vitamin B6 inconjunction with insulin treatment seemed to be better than vitamin B6 or insulintreatment alone in controlling blood glucose, inhibiting platelet aggregation anddecreasing TxA�� production. |
