中文摘要 | ?血營養豐富,含有高量蛋白質,但由於血紅蛋白(hemogloin)具有特殊顏色,故其用途大受限制。本實驗乃利用臭氧之氧化作用將血紅蛋白中之紫質(porphyrin)構造破壞,以期獲得不具色澤之血紅蛋白,並以膠過濾層析法及電泳法探討操作過程中分子量之變化。脫色液之Sephacryl S-400膠過濾層析圖顯示於鹼性(pH 8,9,10)條件下脫色者有高分子量蛋白質生成;而於酸性(pH 2,3,4)條件下脫色者則生成分子量較未處理者低的產物,可見蛋白質分子受到明顯之分解作用,SDS-PAGE電泳圖亦顯示出相同之結果;胺基酸分析結果與硫氫基含量分析結果顯示脫色後已無半胱胺酸與雙硫鍵存在。 |
英文摘要 | The application of swine blood, riched in protein and highly nutritious, was remarkably restricted owing to the unacceptable color of hemoglobin. Ozone, a powerful oxidant, was used to destroy the porphyrin structure in hemoglobin and prepare the decolored protein products. The elution profiles of hemoglobin decolored at basic pHs (pH 8,9,10) on Sephacaryl S-400 showed apparently the formation of higher molecular weight substances; while samples prepared at acidic pHs (pH 2,3,4) showed predominant amounts of lower molecular weight substances. These results indicated the formation and decomposition of proteins, during ozone decoloration, depending on experimental conditions. The patterns of SDS-PAGE also supported the same results. The results of amino acid analysis and sulfhydryl group determination demonstrated that cysteine and disulfide bond were completely destroyed during decoloration process. |
