中文摘要 | 本研究使用懷孕中期女牛的乳腺組織,培養於體外,模擬分娩後乳腺泌乳的狀況,發現乳清蛋白於體外的分泌與乳牛未來的產乳能力有關。乳腺組織以不同濃度的泌乳素處理,在第一天的培養中的α乳白蛋白的濃度很低,而β乳球蛋白卻已經大量存在於培養液內。於第三及第四天的培養中,12.5ng/ml的泌乳素可刺激基本值三倍量的α乳白蛋及β乳球蛋白分泌;400ng/ml的泌乳素則刺激二十倍量的α乳白蛋白分泌,而β乳球蛋白的分泌僅為基本值的四倍。在不同的泌乳素處理中,不論α乳白蛋白或β乳球蛋白,均以第三及第四天分泌最多,之後便開始逐漸下降。於前四天培養所分泌的α乳白蛋白與β乳球蛋白,與分娩後的泌乳量並無相關性;然而第五天及第六天所分泌的α乳白蛋白及β乳球蛋白,卻與分娩後的泌乳量、乳脂量與乳蛋白量呈顯著的正相關,其相關系數約為70%。泌乳素的濃度並不影響期相關性,因此推論在乳腺組織培養後期,一種或多種的非泌乳素因子,可調節α乳白蛋白及β乳球蛋白的分泌,而這個或這些因子對分娩後的泌乳量有關。本實驗顯示,利用懷孕中期乳腺組織培養,可成功預測乳牛未來的產乳能力。 |
英文摘要 | Mammary explants obtained from mid-pregnant heifers were used to mimic the onset of lactation, and the milk protein secretions were correlated to their future lactating ability. Milk proteins produced during the 5th and 6th day of culture were positively correlated with their subsequent milk, fat and protein yields. In the first day of culture, the α-lactalbumin concentrations in media were low in all prolactin treatments, whereas, high concentrations of β-lactoglobulin were found in media. A 3-fold increase from baseline value was found in both α-lactalbumin and β-lactoglobulin sercretions with 12.5 ng/ml prolactin treatment in the third and fourth day of culture. With 400 ng/ml prolactin, α-lactalbumin increased 20-fold, but only a 4-fold increase of β-lactoglobulin was found. The maximun secretions ofα-lactalbumin and β-lactoglobulin in response of prolactin were found in third and fourth day of culture. The secretions of α-lactalbumin and β-lactoglobulin decreased after four days in culture. During the first four days of culture, the sercretions of α-lactalbumin and β-lactoglobulin did not correlate with subsequent milk, fat and protein yields. However, α-lactalbumin and β-lactoglobulin secretions during 5th and 6th day of culture were positively correlated with subsequent milk, fat and protein yields. The correlations are around 0.70. Prolactin did not affect these correlations. These results suggest a prolactin independent factor(s) which regulatesα-lactalbumin and β-lactoglobulin secretion during the later stage of explant culture and has an effect on subsequent milk, fat and protein yields. Therefore, culture of mammary tissue obtained from mid-pregnant heifers can be used for predicting their future lactating ability. |
