中文摘要 | 近年來,本省養雞場爆發馬立克氏病(Marek's disease, MD)的自然病例中,常併發使華氏囊的淋巴球顯著減少的隱孢子蟲症(Cryptosporidiosis),本實驗探討感染隱孢子蟲對雞MD疫苗接種之免疫抑制影響。籍由Sheather's sugar flotation的方法所純化隱孢子蟲的成熟卵囊,經品接種每隻10,000卵囊於1日齡之小雞,並同時皮下接種1劑量的MD疫苗(CV1988株),於接種後第7天和第12天抽血分離血清,與得自MD病雞羽毛囊的MD病毒沉降抗原作用,經由瓊脂沉?反應(Agar gel precipitation, AGP)測定後,接種隱孢子蟲卵囊的實驗組於第7天和第12天的平均AGP抗體力價分別為1倍及0.8倍,而對照組分別為12倍及18.4倍,且經由病理組織切片、電子顯微鏡檢查結果,人工所感染的方式確實寄生存在。由這些結果顯示雞感染隱孢子蟲後,可造成免疫抑制作用,可而可能導致MD疫苗免疫效果失敗原因之一。亦可推測感染隱孢子蟲的雞隻也有較高爆發MD的機會。 |
英文摘要 | For these years, the chickens with Marek's disease (MD) lesions in Taiwan showed the complication of cryptosporidial infection in bursa which caused evident lymphoid depletion. In this experiment, we examined the immunosuppression which were induced by the cryptosporidial infection. The inocula of oocysts were collected from the droppings by purifying with Sheather's sugar flotation method. Two days-old chickens were inoculated by purified inocula of 10,000 oocysts, and vaccinated with one dose of MDV (CVI988 strain) simultaneously. The blood samples were collected for isolating sera, at 7 and 12 days post-inoculation (DPI). The AGP antibodies were tested using cell-free AGP antigens from the follicles of MD chickens. The group of cryptosporidial infection showed the average AGP antibody titers of 1X and 0.8X at 7 and 12 DPI, respectively. By the results of histopathologic and electron microscopic examination, we have establixhed the experimental model of cryptosporidial inoculation. Furthermore, it revealed that cryptosporidial infection caused immunosuppression which resulted the failure of MD vaccination, and showed the high frequency of MD outbreak. |
