中文摘要 | 類前列素(prostacyclin,PG12)與類前凝素(thrombovane,TXA2)為呼吸道及心血 管中重要的類二十碳酸物質 (eicosanoids)。 本實驗以 32 頭一日齡哺乳仔豬分配至 4 個 處理組一分別餵飼 3.6% 椰子油、魚油、大豆油以及豬油,加上 0.4% 大豆油補充各組油脂 的必需脂肪酸,以探討日糧脂肪酸對肺組織與動脈弧生成 PGI2 及 TXA2 之調節機制。每一 處理組中的 4 頭仔豬,於餵飼 2 週後屠宰, 其餘的仔豬則於餵飼 3 過後屠宰,並取肺臟 與動脈弧作組織培養, 培養液以鈣離子泳劑 calcium ionophore( free acid) 作為化學刺 激原,用酵素免疫分析法定量培養液及血清中 PGI2 及 TH2 含量。 日糧脂肪酸組成則以氣 相層析法測定之。結果顯示,餵飼兩週或三週的仔豬,其 PGI2 與 TXA2 的生成改變一致。 魚油組仔豬不但肺臟及動脈弧生成的 PGI2 與 TXA2 低於其他三組 (P<0.05), 血清中濃度 亦較低。鈣離子泳劑顯著地增加動脈弧 PGI2 之生成量,但對肺組織無相同的刺激效果。而 動脈孤 PGI2 的基礎生成量與血清中的濃度成顯著約正相關 (r2=0.53,P=0.007), 顯示動脈 弧內皮組織生成的 PGI2 可能為血清 PGI2 的最主要來源之一。 |
英文摘要 | This study was conducted to investigate the effects of dietary fatty acids on prostanoids released from lung tissue and aorta in neonatal pigs. A total of 32 neonatal pigs from eight litters were fed with diets containing one of four different oils (3.6% coconut oil, fish oil, lard, or soybean oil plus 0.4% soybean oil as essential fatty acid replete) for either two or three weeks. At the end of feeding period, 1 g of lung tissue and 0.5 g of aorta from each piglet were taken and then incubated in 10 ml Tyrode's buffer with or without calcium ionophore at 37 ℃ for 45 minutes. After incubation, prostanoids in 9 ml media and 5 ml serum were then purified by passing through the C18 Sep-Pak column. The amounts ofprostacyclin and thromboxane were determined by measuring their stable metabolites -6-keto-PGF1( (PG) and thromboxane B2 (TX) by enzyme immunoassay. The results revealed consistent changes of the PG and TX synthesis in lung and aorta of piglets feeding for two or three weeks. Calcium ionophore increased PG released from aorta but not from lung tissue. The piglets fed with fish oil diet for two weeks had the lower productions of PG and TX from lung tissue and aorta than other groups (P<0.05). The plasma levels of PG were found to be positively correlated to the basal productions from aorta (r2=0.53, P=0.007). From these results, it was concluded that fish oil containing n-3 fatty acids have the beneficial effect on reducing the risk of vascular diseases by decreasing the production ofprostanoid synthesis. |
