中文摘要 | ?血營養豐富,含有高量蛋白質,但由於血紅蛋白(hemogloin)具有特殊顏色,故其用途大受限制。本實驗乃利用臭氧之氧化作用將血紅蛋白中之紫質(porphyrin)構造破壞,以期獲得不具色澤之血紅蛋白。當調整血液於微酸性(pH 5、6)及中性下進行脫色反應時,蛋白質沉澱量較多,而於pH 4以下或8以上時,則蛋白質回收率高。脫色過程中樣品顏色由紅漸變褐綠,最終可得淡黃色之脫色蛋白液,由分光光度計及色差差之測定可觀察脫色過程中血液顏色之變化,於本實驗條件(通入臭氧量1.17mg/min)下,250ml之血紅蛋白液(蛋白質濃度10mg/ml),脫色時間約五小時。血紅蛋白脫色液經冷凍乾燥製成脫色粉末產品,在外觀顏色上以pH 3者與奶粉較為接近。 |
英文摘要 | The application of swine blood, riched in protein and highly nutritious, was remarkably restricted owing to the unacceptable color of hemoglobin. Ozone, a powerful oxidant, was used to destroy the porphyrin structure in hemoglobin and prepare the decolored protein products. Hemoglobin precipitated obviously when ozone decoloration was conducted at weak acidic pHs (pH 5 or 6) and neutral pH, but higher protein recoveries were obtained when ozone treatments were carried out a pHs below 4 or above 8. Spectrophotometer and color difference meter were employed to record and to compare the color changes of sample solutions. In appearance, the lyophilized decolored-hemoglobin, prepared at pH 3, was much resemble to milk powder. |
