| 中文摘要 | 本試驗之目的,係探討本省飼養的巴貝多黑肚綿羊與臺灣黑色土山羊之各季節之超級排卵之差異,並探討簡化超級排卵之方法,以供基因轉置動物生產,和卵和胚顯微操作研究之用。將24頭黑肚綿羊和36頭山羊各逢機分配於一年四季,綿羊群放置助孕素塞劑(CIDR)於陰道內10日後取出,於取出CIDR前48小時開始每日2次遞減之量分3.5日肌注16mg之pFSH,羊隻於CIDR取出後54小時發情,加以配種,配種後第6日以外科手術採胚,並計算黃體數,山羊則不使用CIDR,僅於發情後第9日開始遞減量3.5日注射16mg pFSH。同時,綿羊與山羊各12隻,逢機各被分成2群,一群羊隻注射16mg pFSH溶於30% polyvinylpyrrolidone (PVP),僅進行一次注射,另一組對照組以遞減量分3.5日7次注射。試驗結果綿羊與山羊春夏秋冬各季節採胚時黃體數分別為14、17、17、17個和9、7、8、8個。採到的胚數分別為12、9、6、8個和4、3、4、4個,其中可移置胚數分別為9、5、4、8個和4、3、3、4個。雖然綿羊於春季可採取較多的可移置胚數,但黃體數並無較多。山羊則春季黃體較多,但可移置胚數並無增多。本試驗結果證實黑肚綿羊與本省土山羊於本省南部環境下均可採到試驗用胚。而以pFSH-PVP一次肌注之簡化試驗,於綿羊之反應較佳,可得6頭黃體,但比多次注射組黃體少一半。山羊之反應頭數及黃體數均減少。 |
| 英文摘要 | The objective of this study was to compare the superovulatory response of Barbados black belly sheep and Taiwan Native Goat in the various seasons with a single pFSH-PVP injection. Twenty-four ewes and 36 does were sampled at random during the four seasons. Ewes were treated with an intravaginal device called the control internal drug release (CIDR) for 10 days and treated with a 3.5 days declining dose of 16 mg (3.3, 2.2, 2.2, and 2 mg) injections every 12 h, beginning at 48 h before CIDR removal. Does were treated with 16 mg pFSH beginning at 9 days after natural heat, and received 125 ug i.m. of cloprostenol. (Estrumate) at 48 h after pFSH injections. In the meantime, 12 ewes and 12 does were divided into two groups and treated with 16 mg pFSH in a single injection and in multiple declining dose injections. Ewes and does were hand mated three times in estrus, and the embryos were collected on day 5~6 after estrus and the corpora lutea were calculated. The numbers of corpora lutea obtained in spring, summer, autumn, and winter averaged 14, 17.6, 17 and 17.7, respectively in sheep and 9.3, 7.4, 7.9 and 7.5 respectively, in goat. The numbers of transferable embryos recovered from each ewe of black belly sheep and native black goat were 5.9 and 3.5, respectively. Numbers of transferable embryos recovered in spring, summer, atumn, and winter seasons were 9.2, 5.2, 4.2 and 5, respectively, from superovulated sheep; and those from native black goat were 4.4, 3.3, 2.4 and 3.8, respectively. The numbers of corpora lutea detected from the superovulatory response by a single injection of pFSH dissolved in PVP were smaller than for the group of multiple pFSH injection (6 vs 14, 3 vs 8) in Barbados black belly sheep and native goat using 16 mg pFSH. The results indicated that higher superovulatory response can be obtained throughout the whole year not only in black belly sheep but also in native black goat. |
