中文摘要 | 冷凍保護劑對於九孔卵滲透速率的量測與模式分析,可提供建立其低溫 保存程序所需之重要資訊,亦可以縮短建立冷凍保存程序之實驗流程。本研究中 應用微擴散室系統分別量測得五種冷凍保護劑對九孔卵滲透過程中所產生之體 積變化資料,從而比較不同冷凍保護劑之滲透速率。對於九孔卵而言,冷凍保證 劑在20℃之滲透速率由高至低依序為:丙二醇 >二甲亞�� >乙醯胺 >乙二醇,甘 油對九孔卵幾乎不滲入。而冷凍保護劑對於九孔卵的滲透現象,則以不可逆熱力 學為基礎建立數學模式,並以非線性迴歸方法分析丙二醇滲入九孔卵過程所量測 得之體積變化數據,以量化丙二醇對九孔卵滲透的動態行為,分別求得在10、20 與30℃時丙二醇對九孔卵之水滲透係數Lp、可滲透溶質移動力ω與反射係數σ。 所求出的滲透模式參數組可用於進行電腦模擬,以預測不同濃度與溫度狀態下, 添加或移除冷凍保護劑過程中卵之體積變化,以及卵內濃度之變化,據以建議添 加冷凍保護劑於九孔卵之最佳溫度、濃度與時間。另外,電腦模擬的結果亦可用 以解釋與預測於移除冷凍保護劑過程中,添加非滲透性溶質以得到滲透壓緩衝的 效果。 |
英文摘要 | In order to establish a successful cryopreservation protocol forsmall abalone eggs, it is necessary to characterize and model thecryoprotectant permeation process to the eggs. This will not onlyprovide valuable information but also avoid unnecessary trial anderror experiments in designing the cryopreservation protocol. Amicro-diffusion chamber system was used to measure the volumetricexcursion of small abalone eggs when they were subject to variousconditions of cryoprotectant permeation. Five cryoprotectants weretested and their permeation rates were compared based on thevolumetric data. The order of permeability for small abalone eggs at 20 ℃ was propylene glycol > DMSO > acetamide > ethylene glycol.Glycerol appeared to be nonpermeable to small abalone eggs. Amathematical model was established based on irreversiblethermodynamics to describe the cryoprotectant permeation process tosmall abalone eggs. The volumetric data of propylene glycolpermeation at 10, 20 and 30 ℃ were fitted with the mathematicalmodel using nonlinear regression to determined the model parametersLp ω and σ . Computer simulation were performed with theestimated parameters to provide information which can be used tosuggest selection of appropriate temperature, timing andconcentration during loading or unloading of cryoprotectants. Thesimulation results also revealed the effect of adding nonpermeatingsolute as osmotic buffer during unloading of cryoprotectant fromsmall abalone eggs. |
