中文摘要 | 桔西氏錐蟲(Trypanosoma cruzi) 及臺灣蝙蝠錐蟲(Trypanosoma taiwanianum)是 在半翅目 (Hemiprera) 的昆蟲宿住及恆溫的脊椎動物宿主之間交替循環生活。 在此複雜的 生活史中, 錐蟲至少具有四種不同形態: 側鞭型 (epimastigote) 、 錐體型 (trypomastigote)、圓鞭型 (spheromastigote) 及無鞭型 (amastigote)。將臺灣蝙蝠錐蟲 以 37 ℃培養 2 小時,然後再以 41 ℃培養 1 或 2 小時後, 其圓鞭型及無鞭型有增加的 趨勢。 培養之臺灣蝙蝠錐蟲無論是對照組或是以熱休克處理者, 皆可出現少數錐體型蟲體 (<1.3%),而以同樣培養基培養之桔西氏錐蟲則無此種形態。 經過熱休克處理之桔西氏錐蟲 在試管底部有蟲體聚集現象,以 41 ℃處理 1 小時至 2 小時後,側鞭型蟲體出現之百分比 由 70% 增加至 87%,以 37 ℃培養 2 小時再以 41 ℃培養 1 至 2 小時後,側鞭型蟲體出 現之百分比由 91% 增加至 99%。經十二烷硫酸納 - 聚丙烯胺膠體電泳 (SDS-PAGE) 對兩種 錐蟲經熱休克處理後蛋白質分析,在溫度升高時,大多數蛋白質的合成受抑制。經過熱休克 處理後有些類似熱休克蛋白質 (heat shock protein) 的白帶出現,在桔西氏錐蟲有分子量 為 70.5 及 74.9kDa 的蛋白質,而臺灣蝙蝠錐蟲蛋白質的分子量為 53.6,69.6 及 70.4kDa 。 |
英文摘要 | Bothe Trypanosoma cruziand Trypanosoma taiwanianum display complex life cycles involving two hosts, a hemiptern insect and a vertebrate. During the development of their life cycles the parasites exhibit at least four different forms:epimastigotes, trypomastigotes, spheromastigotes and amastigotes. It is shown in our study that high temperature stimulates the increase of amastigote and spheromastigote forms in T. taiwanianum if it was heat-treated at 37 ℃ for 2h and then raised to 41 ℃ for 1h or 2h. Trypomastigotes existed at 27 ℃ in the control group and other heat-treated groups in T. taiwanianum (<1.3%), whereas they could not be found in T. cruzi. It revealed that these two species of trypanosomes have different morphological compositions even in the same culture medium. Pellets of organisms were found at the bottom of test tubes after heat treatment in T. cruzi. The percentage of epimastigotes increased with the periods of heat shock: 70% when placed at 41 ℃ for 1h increased to 87% at 41 ℃ for 2h. When placed at 37 ℃ for 2h and then raised to 41 ℃ for 1h. The percentage of epimastigotes was 91%, which increased to 99% when placed at 37 ℃ for 2h. then raised to 41 ℃ for 2h. In comparison to the heat shock proteins induced in T. cruzi and T. taiwanianum as analysed by Sodium doedcyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), most protein syntheses were inhibited at the elevated temperature. However, several bands resemble HSPs were induced, 70.5 and 74.9 kDa in T. cruzi, 53.6, 69.6 and 70.4 kDa in T. taiwanianum. |
