中文摘要 | 本試驗所用材料係以代表泡桐屬3個組之模式種:泡桐(P. fortunei) ,白桐(P. kawakamii)與毛泡桐(P. tomentosa)之培植體,培養在不同的的培養基上,以探討其器官發生能力之差異及發展適合所有泡桐屬的再生培養基之可行性。台灣泡桐(P. xtaiwaniana為泡桐與白桐的雜交種,且為省產重要經濟樹種故亦一起參與試驗。所用的培植體為:子葉、下胚軸、頂芽、節間、節與葉片。培養基為MS基本培養基添加各種不同組和的 NAA與BA。MS培養基添加0.5mg/l的NAA與5mg/l的BA均能使所有樹種與培植體產生不定芽。不過樹種與培植體的來源對器官發生力有相當大的差異:四個樹種的下胚軸與子葉及毛泡桐與台灣泡桐的所有的培植體,其不定芽僅發生在特定組合的NAA與BA(0.5+5,2.5+1及2.5+ 5mg/l);而泡桐與白桐的預芽、節、節間與葉片培植體則可在大部份的NAA與BA組合中誘導出不定芽。不定芽之發生經組織切片顯示有直接與間接發生二種,直接發生係從培植體表皮或皮層細胞直接產生,間接發生則由切口處之癒傷組織發生。不同的凝膠質(水昌凝膠gelrite及不同商品的洋菜粉Difco或Biolab agar)對不定芽的形成率、癒傷組織的生長與玻璃質化芽的產生均有影響。毛泡桐對凝膠質的種類在不定芽的形成與癒傷組織之生長甚為敏感。在所用的凝膠質中,以0.8%Difco agar可誘導較穩定之不定芽之產生及減少癒傷組織的生長。 |
英文摘要 | Organogenesis of three sections of Paulownia genus: P. fortunei, P. kawakamii and P. tomentosa were examined in six types of explants: cotyledon, hypocotyl, shoot tip, node, internode and leaf. P. taiwaniana, an economically important tree which was proved a hybrid between P. fortunei and P. kawakamii, was also included for conparison. Explants were cultured on MS medium supplemented with various combinations of NAA and BA. Medium containing 0.5 mg/l NAA and 5.0 mg/l BA induced adventitious buds in all explants for all four species. However, organogenetic capacity depends on explant types and species. For cotyldeon and hypocotyl explants of four species and all explants of P. tomentosa and P. kawakamii formed buds on most combinations of NAA and BA. Hsitological study revealed that adventitous bud formation occurred indirectly from the callus which derived from cutting surfaces of explants or directly from epidermis or cortex cells. Gelling agents affected adventitious bud formation of explants, callus growth and vitrification of shoots. P. tomentosa was very sensitive to different gelling agents in adventitious bud formation and callus growth. Difco agar at 0.8% concentration gave explants a stable induction of adventitious buds and less growth of calllus. |
