中文摘要 | 基因工程生產的豬生長素,溶入緩衝液再以生理食鹽水稀釋後,便用輸液馬達依 1/μg/kg/h的劑量,持續注入60公斤閹公豬的靜脈內。此種處理係模擬緩釋激素之作用, 來探討持續外源生長素的刺激,對血中生理性生長素波動的影響。靜脈灌注的期間為晚間8 小時,灌注期4小時先以空白灌注作為對照。血中一些相關的代謝物和胰島素亦一併測定, 以觀察可能引起的變化。 試驗結果顯示顯示,灌注期內的生長素波峰數比對照期每小時平均少0.1次(0.43 v. 0.53);生長素基底濃度降低68%(0.93 v. 1.37 ng/ml),但最高波峰值增加15%(4.84 v. 4.20 ng/ml), 然而這些差異均未達統計顯著水準,從基低濃度至最高值的倍數來考量,處理期間 內是5.20倍,而對照期為3.13倍,而這是因灌注期基底值較低的結果。以上的資料顯示, 本試驗的生長素灌注,並未影響生長素的波動輪廓。豬隻血中的尿素和葡萄濃度則顯著地 (P<0.05)增加,這也表示生長素的確造成了代謝上的影響。 本試驗的結果,可獲一結論,即低劑量如1/μg/kg/h的生長素灌注,對閹公豬血中,生 長素濃度的生理素波動,不會產生顯著的影響。國外施用緩釋生長素所得的結果,因其為專 利裝置,作用模式不能確知,故與本試驗的結果,不具比較意義。 |
英文摘要 | Recombinant porcine somatotrophin (rPST), diluted with buffer/saline, was infused (1μp/kg/h) into 10 barrows for 8h during the night to simulate the sustain release of the hormone. Blank infusion was conducted prior to the introduction of the hormone for 4h as control. The changes of physiological profile of circulating somatotrophin, some blood meta-balites and insulin level due to the infusion were investigated. Results showed that, during rPST infusion, the number of hormone release peak was in average 0.1/h less and the basal level was 68% of the control. However, the maximum level of peaks was 15% higher than in blank period and resulting a greater elevation ratio of 5.20 compare to 3.13. Those data indicating that rPST treatment in this study exerted little effect on circulating somatotrophin profile in growing barrows. The blood urea and glucose concentrations were not altered by the infusion, but the insulin level was increased by 15% (P<0.1) and the free fatty acid content was elevated significantly (P<0.05). These data demonstrated that the rPST infused did has a metabolic effect. It is concluded that rPST infusion at the rate of 1μp/kg/h could not change the physiological profile of the hormone. Comparasion of the present results with references using sustain release devise of rPST will be meaningless since the release pattern of the devises has not been revealed. |
