中文摘要 | 為瞭解產黃纖維單胞桿菌(Cellulomonas flavigena)NTOU1株產生的色素對利用本菌生長的蝦醬油的食品安全與營養評估的參考,進行本試驗。本試驗以無機鹽培養基(M70)為基礎培養基;以電子顯微鏡觀察本菌在不同營養下的形態變化;以薄層分析、質譜儀,並參考C.biazotea的結果鑑定色素。本C.flavigena NTOU1株在MG70(M70 + O.1,w/v,葡萄糖)培養基中能生長,但不產生色素,甚至於含維生素B1,或生物素也不產生色素。然在MG70培養基含維生素B1,及生物素、或維生素B1及組織胺、或先加維生素B1,培養48小時後再加白胺酸,在72小時內均會產生黃色色素。本菌生長或產生色素之最適pH為7.0-9.0或9.0;它略能利用甘露蜜糖和棉子糖,但最能利用纖維二糖、麥芽糖及木質糖以生長及產生色素。生物素能保持本菌在生長時呈正常桿狀,但形狀與色素產生無關,所產生的色素似類胡蘿蔔素,因此它在食品加工上應無營養及安全上的顧慮。 |
英文摘要 | Exogenous factors for growth and pigmentation of Cellulomonas flavigena NTOU I were studied using mineral salts (M70) medium as a base. Morphological changes of the bacterium in the media containing different nutrients wereobserved under transmission electron microscope. Yellow pigment was identifiedusing thin-layer chromatography and mass spectrum, comparing with C. biazoteadocumented. The bacterium could grow but could not produce pigment in MG70 medium (M70 medium +1.0 glucose), even if thiamine or biotin was present inthe medium. The bacterium was pigmented when cultivated in MG70 mediumcontaining thiamine and biotin, thiamine and histidine, or thiamine and leucinewhich was added 48 hr after inoculation during 72 hr incubation. The ptimal pHfor growth of the bacterium was 7.0-9.0, while pH 9.0 was favor for pigmentproduction. As a carbon source, mannitol and raffinose were not available forthe bacterium. The bacterium could effectively utilize cellobiose, maltose andxylose for growth and pigmentation. Biotin played an important role on keepingregular rod-shape of the bacterium. Morphology of the bacterium is little relation to its pigmentation. The yellow pigment may be a mixture of carotenoids. |
