中文摘要 | 佐劑可以幫助抗原提升較早,較高和較長的免疫反應,所以利用佐劑可以減少抗 原的需要量,降低生產疫苗的成本。細胞培養產製日本腦炎疫苗加入佐劑例如:磷酸鋁、氫氧 化鋁、磷酸鈣及BCG皆能有效的增加anti-E3抗體的量和中和病毒的能力。Montanide*ISA是 一種水溶於油(W/O)佐劑,此佐劑無法有效增高疫苗效價。利用酵素連結免疫反應法測定老鼠 免疫血清中anti-E3抗體的量,在pH值7.4左右,磷酸鋁增加anti-E3抗體量的效果優於氫 氧化鋁。另外兩種鋁鹽佐劑不論是增加anti-E3抗體量或是中和病毒的能力都是以劑量為 0.6mg/mL時的效果最好。在相同鋁鹽佐劑劑量下,0.4mg/mL磷酸鋁,pH值為7.5時,anti-E3 抗體產生的量最高,但中和病毒的能力則以pH值為7時最高。然而磷酸鋁在0.8mg/mL,pH 值為7.5時免疫效果最好。 |
英文摘要 | Adjuvants are used for conventional vaccines to elicit earlier, higher and/or longer lasting immune responses. Optimal application of adjuvants will thus reduce the amount of antigen required, thereby enhance safety and reduce production cost. Our study shows Japanese encephalitis vaccine in the presence of adjuvant like aluminum phosphate, aluminum hydroxide, calcium phosphate, or BCG, with the exception of Montanide* ISA, markedly induces higher titer of antigen-specific antibody (anti-JE-E3) and plaque reduction neutralization test (PRNT) titer. Comparing antibody concentration and PRNT titres, our results indicate that aluminum phosphate can induce an immune response much better than aluminum hydroxid. We have found the optimal dosage of aluminum phosphate or aluminum hydroxide for Japanese encephalitis vaccine is around 0.6mg/mL, and we recomomend that aluminum phosphate in the range form 0.4 mg/mL to 0.8 mg/mL with the optimal pH around 7-7.5 is used as an adjuvant for Japanese encephalitis vaccine. |
