中文摘要 | 運用單株抗體合併免疫螢光反應,我們在雞胚水晶體早期發育的階段, 定位出鹽基性纖維芽細胞成長因子(bFGF)在雞胚第四天時,bFGF即在水晶體囊 (lenScapsule)發現,而在雞胚第十天時,其免疫組織反應下降。但反而在水晶體 上皮細胞有顯看反應,並發現原發水晶體纖維(primarylensfiber'.已發育完成,而且 免疫反應在上皮細胞各層均勻分佈。 由本實驗,我們推論bFGF在早期水晶體發生中扮演重要角色。近來水晶體中的 bFGF來源有兩派假說,有一派假說認為bFGF是由玻璃體或房水而來(Paracrine), 另一派假說則認為bFGF是由水晶體上皮細胞本身產生(Autocrine),但至今尚未有 結論。本研究先在水晶體囊發現bFGF,繼之又在水晶體上皮細胞發現有更顯看 的bFGF反應,或可推論由Paracrine來的bFGF較由Autocrine來的bFGF較早對胚胎之 水晶體發育產生影響。 |
英文摘要 | Purpose: We localized basic fibroblast growth factor (bFGF) in chickembryo lens so as to specify its role in early lens development. Methods: By means of monoclonal anti-bFGF antibody in conjunctionwith immunofluorescent techniques, chick embryo eyes from 4 days ofincubation (E4) to 7 days of incubation (E7) were used for study. Results: The bFGF was detected first in lens capsule by E4. At E7, itsimmunoreactivity in capsule decreased, however, lens epithelium showedconspicuous reaction instead, accompanied with full development of primarylens fibers. The bFGF reactivity was shown to be evenly distributed invarious layers of lens epithelium. Discussion: The temporal and spatial expression of bFGF in embryoniclens indicates an important role for bFGF in early lens morphogenesis.Recently, the origin of bFGF in lens has been proposed as either fromvitreous and aqueous (paracrine) or produced by lens epithelial cell itself(autocrine). In this study, the first presence of bFGF in lens capsule whenthere is no detectable reaction in epithelium, followed by its appearance inwhole epithelial layer later, raised the possibility that paracrine-origin bFGFmay exert its influence sooner than autocrine-origin bFGF. |
