中文摘要 | 取濕地松60~80天苗齡之實生苗作為培殖體,使用cytokinin以浸泡或添加在培養基中的方式誘導其側芽抽長進行微體繁殖。不論採用的是何種誘導方式,培殖體維持全株完整、不要截成多段比較能產生多量的芽體。以短暫浸泡作為誘導處理時,6-benzylaminopurine (BAP)和kinetin (KT)彼此間無法區分出何者較佳。但是高濃度BAP (100 mg/l)會抑制芽體抽長。在GD-1培養基中添加1 mg/l BAP作為誘導側芽生長之環境時,以處理30~45天即能達到最佳之誘導效應。在本研究中芽體產出情形最佳的處理,為不截段的培殖體以1.0 mg/l BAP、添加在培養基中的方式誘導四週,再移往含活性碳的培養基中四週。平均一株實生苗在經過八週的誘導與繼代培養之後,可產生3.31個大芽(長度>3.0 mm),4.19個小芽(長度≦3.0 mm)。 |
英文摘要 | Slash pine seedlings grown 60-80 days after germination were collected as explants in this study. Cytokinin application was carried either by temporary immersion or by medium addition in order to stimulate the axillary bud elongation. N matter what kind of stimulation the seedling explants received, they would produce more axillary bud from intact or less segmented specimens. 6-benzylaminopurine(BAP) and kinetin(KT) were equally effective to induce bud production when using immersion method. But BAP at a higher concentration (100 mg/1) inhibited bud elongation. When using medium addition method (BAP 1 mg/1 in GD-1 medium), the better treatment duration was determined at about 30-45 days. We have had a best bud induction treatment in this study that a seedling explant would well response to the BAP (1.0 mg/1) treatment through a 4-week medium addition method, and then cultured with charcoal (minus BAP) for 4 more weeks. This procedure produced 3.31 elongated large buds (>3.0 mm) and 4.19 non-elongated small buds (≦3.0 mm) per explant. |
