中文摘要 | 本研究之目的,在於嘗試開創雄鵝精液之冷凍保存技術,俾提供作為鵝 人工授精之使用。供冷凍雄鵝精液用之溶液有二:一為在Lake's溶液中添加10之甘 油(glycerol),另一則在Beltsville Poultry Semen Extender(BPSE)中添加4DMSO之BPSE 作為冷凍溶液,並以二階段式降溫法,將雄鵝精液以-1℃/min.之速度先自25℃ 降溫至-35℃,再移入液態氮中予以冷凍保存者,經取出並於0解凍後,證明精子 之活力與受精率,便分別為.7與0。在相同冷凍與解凍條件下,將冷凍溶液改用 Lake's溶液添加10甘油為之,則精子經冷凍暨解凍後其活力(28.4)與受精率(9.7), 分別均顯著優於應用含4DMSO之BPSF作為冷凍溶液者(P<0.01)。尤有甚者,改用 二階段降溫法,將懸浮於Lakp's溶液中之雄鵝精液,先於5℃開始以-1℃/min.之 速度降至-20℃,再使用-30℃/min.之降溫速度,使溫度降低至-80℃,並維持l0min. 後,再移至液態氮中予以冷凍保存者,經使用0℃之解凍溫度予以解凍並舉行人 工授精,結果可獲得31.6之受精率;提高解凍溫度至41℃者,更使其受精率提高至 38.5。 |
英文摘要 | An attempt was made in this study to establish an effectivefreezing system for long term presevation for gander semen in theliquid nitrogen (LNg) in order to meet the requirement using for theartificial insemination. Two extenders, one was the Lake's solutionsupplemented with 10 glycerol and the other Beltsville poultry semen extender (BPSE) supplemented with 4 dimethylsulfoxide(DMSO), were used for freezing the gander semen. Gander semenafter suspended respectively in the said extenders and subjected forfreezing with a cooling rate set at -l℃/min from 25℃ to -35℃followed by pluging them into the LN��. Frozen semen were thenthawed at 0℃ for the evaluation of the sperm motility and thierfertilizing capacity. Results showed that by using BPSE solution as the frozenextender, only 9.7 of the frozen-thawed sperm were progressivelymotile and virtually no egg was fertilized when the frozen-thawedsemen was used for Al. In place of the BPSE, when the Lake'ssolution was used for freezing gander semen, much higher proportion(28.4) of sperm appeared their progressive motility after they hadbeen frozen and thawed under the same conditions and 9.7 of eggscollected within one week post insemination with the frozen-thowedsemen was fertilized. Moreover, changes of the freezing programme by cooling at -1℃/min from 5℃to -20 ℃ and further cooling at -30℃/min from -20℃ to-80℃, holding at -80℃ for 10 min before the frozen semen had beenplunged into the LN��, the fretilization rate was further increased to31.6 when the frozen semen had been thawed at 0 ℃ andsubsequently used for AI. However, the optimal temperature forthawing these frozen semen was 41℃ which resulted in 38.5 of eggsfertilized after insemination the frozen-thawed semen to geese. |
