中文摘要 | 肝細胞是氯基酸代謝主要場所之一,切肝術後之再生應與氯基酸之供應關係密切;而側鏈氯基酸與肝細胞再生速度也可能有關。本實驗以重約200公克之雄性Wistar大鼠,分為三組,每組32隻,各切除其肝臟之中間葉及左側葉(共67.31%),再分別給予高濃度氯基酸輸液(HA組)、高濃度側鏈氯基酸輸液(HB組)、及口攝Chow 50001飲食(H-O組),另一組大鼠(32隻)只作剖腹假性手術,術後亦給予口攝飲食(S-O組)。各組大鼠分別在術後6、24、48、72小時犧牲取樣,測量及比較其剩餘肝臟DNA總含量,並比較分析各組大鼠血清氯基酸及肝功能測定的改變,結果發現(1)肝臟部份切除後72小時內,血清氯基酸均明顯下降,尤以側鏈氯基酸為甚;(2)給予高濃度氯基酸輸液之大鼠,其術筱剩餘肝臟內去氧核醣核酸(DNA)總含量沒有比H-O組多;(3)給予高濃度側鏈氯基酸輸液之大鼠,術後72小時之剩餘肝臟內DNA總含量,則明顯比H-O組及HA組高。 |
英文摘要 | Through a variety of anabolic and catabolic processes, the hepatocyte is one of the major sites of amino acid metabolism. In general, amino acids are utilized for the synthesis of liver intracellular proteins. It has been suggested that amino acids play an important role in liver regeneration after partial hepatectomy. Branched-chain amino acids (BCAA) are also mentioned when studying the factors affecting liver regeneration. This study evaluated the effects of amino acids or BCAA on the liver regeneration, and analyzed the serum amino acids level after partial hepatectomy in rats. Male Wistar rats weighing around 200g were used as subjects. Partial hepatectomy with resection of the median and left lateral 10bes (67.31%) was performed. High amino acid (HA) or high branched-chain amino acid (HB) solutions were given intravenously two days prior to the partial hepatectomy. The rats were sacrificed at 6, 24, 48, 72 hours after operation. Remnant liver deoxyribonucleic acid (DNA) content was chosen as marker for comparing the effects on liver regeneration. The results were: (1) Almost all amino acids, especially BCAA, decreased in serum concentration within 72 hours after partial hepatectomy; (2) HA group rats showed no higher DNA content in the remnant of liver than H-O group rats 72 hours postoperatively; (3) HB group rats showed a significant higher DNA content in the remnant of liver than H-O and H-A groups rats 72 hours postoperative1y. |
