中文摘要 | 銀柳材料的消毒,以1.0%次氯酸鈉溶液添加0.1%Tween20,經由真空抽氣15分鐘的 效果較佳。滅菌後的銀柳枝條切體置入MS培養基(Murashige & Skoog's medium,1962)添 加Sucrose 3%,Agar 0.8%,pH=5.8及不同組合及濃度之植物荷爾蒙做組織培養後發現,不 同處理中以1ppm 2,4-D具有較佳之癒傷組織誘導效果,而3ppm及5ppm 2,4-D所誘導之癒傷 組織則太過膨鬆。 經由2,4-D所誘導之癒傷組織再移入Anderson's medium(1984)中,添加3% Sucrose, 0.8% Agar及不同植物荷爾蒙作不定根及芽體誘導試驗,結果發現不定根的形成以2ppm IBA 之效果最佳,而芽體的形成則以0.5ppm BA或2ppm Kinetin之效果最好。 |
英文摘要 | The best procedure for sterilization of the explants of Cat Tail Willow was surface disinfected in a 1.0% NaOC1 solution containing 0.1% Tween 20 with vacuum-pump-letting for fifteen minutes. Explants were cultured on MS medium (Murashige & Skoog, 1962) supplemented with 3% sucrose, 0.8% agar and different kinds and concentrations of plant hormones. The pH was adjusted to 5.8 with 1N NaOH or 1N HCI prior to autoclave. The result shows that 1ppm of 2,4D- is more effective than 3 or 5ppm in inducing of callus formation. The callus induced by 2,4-D were transplanted to Anderson's medium(1984) supplemented with 3% sucrose, 0.8% agar and various concentrations of plant growth regulators for inducing of shoots and roots formation. The results indicated that for 2ppm IBA, and 0.5ppm BA and 2ppm Kinetin had the best effects in inducing of shoot and root formation, respectively. |
