| 中文摘要 | 本實驗目的在探討大白鼠長期處於噪音情況下,是否會影響其血壓及血管反應性。大白鼠經一、二、三及四星期噪音(100分貝、1000赫、4小時/天)處理後,以夾尾(tail-cuff)法測其血壓並以二及四星期之離體胸主體脈測試其對各種血管收縮劑和舒張劑的反應。結果顯示經一或二星期噪音處理後,動物血壓並無明顯變化,但經三或四星期噪音處理後則動物血壓顯著增加(噪音前:110±5 mmHg,噪音後;128±5 mmHg,141±6 mmHg)。此外,離體胸主動脈血管的反應性;噪音會引起血管對serotonin(5-HT)收縮反應增強,但phenylephrine(PE)及高鉀則無明顯變化。對內皮依賴性血管舒張劑如acetylcholine(Ach)及A23187舒張反應在噪音處理組顯著小於對照組,且其對Ach的舒張反應可完全被methylene blue(MB, 10-5 M)及Nω-nitro-L-arginine(LNNA, 5×10-5M)阻斷;而對非皮內依賴性血管舒張劑nitroglycerin(NG)的舒張反應則無明顯差異。本實驗結果顯示動物長期處於噪音情況下會使血壓上升,而引起此血壓上升之可能原因部份是由於噪音影響血管內皮功能改變所致。 |
| 英文摘要 | The present study was designed to determine whether there is a causal relationship between noise-induced hypertension and changes of the endothelial function. Rats were exposed to noise stress (100 dB, 1 KHZ, 4-hr/day)for 1-4 weeks. The systolic blood pressure was significantly increased after rats exposed to noise for 3 or 4weeks but not 1 or 2 weeks. As results demonstrated in isolated thoracic aortic rings, the responses to serotonin (5-HT) were enhanced in noise-treated rats while there were no changes in response to phenylephrine (PE) and high-k+ between noise-treated and control rats. The relaxant to endothelium-dependent vasodilators-A23187 and acetylcholine (Ach) in noise-treated rats were less than those in control rats. The vasodilator responses to acetylcholine were completely abolished by methylene blue (MB) or Nω-nitro-L-arginine (LNNA). Whereas, the responses to endothelium-independent vasodilator, nitroglycerin (NG), were not significantly changed after rats exposed to noise. To summarize our results, the enhancement to vasoconstrictors and the attenuation to endothelium-dependent vasodilators may account for elevation in blood pressure during noise stress. This indicates that the elevation in blood pressure by noise stress may be partly due to the deterioration of endothelial function. |
