中文摘要 | 以0、0.2、0.4及0.8mM methanesulfonic acid methyl ester (MMS)處理中國倉 鼠卵巢細胞(Chinese hamster ovary cells,CHO-K1),並以Alkaline elution技術測定CHO- K1之DNA單股斷裂(single-strand-break,SSB)之情形。結果顯示:添加0.2mM以上之 MMS,於elution 2 hr,其retention ratio便顯著小於未處理組(0.47 vs 0.96),亦即0.2mM MMS便可顯著引起DNA SSB,讓DNA發生SSB之程度與所添加MMS之濃度成正相 關,且隨elution之時間而增加其嚴重性。Alkaline elution技術為研究細胞中DNA單 股斷裂之理想方法。 |
英文摘要 | Treating Chinese hamster ovary cells ( CHO-K1 ) with 0, 0.2, 0.4 and0.8 mM methanesulfonic acid methyl ester ( MMS ) , and testing the amountof DNA single-strand-break ( SSB ) by alkaline elution technique. The resultsshowed the retention ratio of DNA were decreased significantly treated with0.2 mM MMS and inversely with the concentration of MMS added and theamount of DNA SSB was increased linearly with the elution time. Alkalineelution analysis is a reasonable method to study DNA single-strand-breakage. |
