Meyer)不定根組織於含有0.5/mg/l 2,4-D (2,4-dichlorophenexyacetic acid)及0.1 mg/l 6-BA(6-benzyladenine)之MS培養基,可誘導形成癒合組織及不定根,而其中的癒合組織再經繼代培養於含有1 mg/l 2,4-D之MS培養基,可形成體胚。此成熟體胚置於MS基本培養基中發芽長成之幼株,再培養於含有2 mg/l IBA之1/2 MS培養基,有65%之幼株可發根。再生三小葉片幼株由培養容器移至大氣中之土壤與蛭石混合介質中,經健化後,外觀和實生三葉片幼苗相似。 |
英文摘要 | Cross sections of the adventitious root of ginseng (Panax ginseng C. A. Mayer) were induced to form calli with 95% efficiency on MS medium supplemented with 0.5 mg/l 2,4-D and 0.1 mg/l 6-BA. These calli when cultured on MS medium enriched with 1 mg/l 2,4-D produced numerous embryoids which able to form individual shoots and multishoots with poor root systems in basal MS medium. A 65% rooting efficient process was established for the individual shoots culturing in 1/2 MS medium supplemented with 2 mg/l IBA. The regenerated plantlets were transplanted from the culture tubes to the vermiculite-soil mixture and further grew into three-leaflet plants with 60% efficiency four weeks after culturing under ambient environmental conditions. The results showed that unfolded three-leaflet plants could be regenerated from the calli of adventious roots within fourteen months. |
