| 中文摘要 | 用螢光染劑fluorescein isothiocyanate (FITC)及rhodamine isothiocyanate (RITC)來標誌甘蔗(F 177品種)懸浮培養細胞。22小時後,將一部份標誌的細胞用培養液清洗,另一部份則不清洗。清洗過的細胞也分兩部份:(1)供作生長觀察;(2)供作原生質體分離之用。所有細胞之生長速率都每兩天測量一次,直至第十天。總合清洗細胞及不清洗細胞之資料可知:FITC,RITC及酒精(溶劑)處理過的細胞,其生長速率與對照(正常細胞)者無顯著差異。清洗過的細胞之生長速率,雖然在培養十天後超過不清洗者,但兩者之差異尚未達百分之五顯著水準。螢光劑標誌之細胞,其原生質體分離率超過98%。將分離後的原生質體培養於C₈IV液體培養基中,17天後,FITC及RITC標誌之原生質體,其細胞團形成率與對照(酒精)者無差異,可見外來螢光劑對原生質體之分裂及生長無毒害作用。更有進者,此時由RITC標誌之原生質體所生成的細胞團仍呈淡紅色。此一長時間不褪色的特性,可作為一種標誌,用以選拔真正體細胞雜種。 |
| 英文摘要 | Fluorescein isothiocyanate (FITC) and rhodamine isothiocyanate (RITC) were used to prelabel suspension cells of sugarcane (Saccharum spp. hybrids, cv. F177). After 22 hr, one portion of the cells was washed, the other was not. Part of the washed cells served as the source for protoplast isolation. The growth rates of all types of cells were measured every other day until the 10th day. The FITC-, RITC- and alcohol (EtOH)-treated cells grew as rapidly as the normal cell (control) before the first eight days. Their differences became larger at the 10th day but still didn't reach the 0.05 probability level. This phenomenon is in agreement with the following experimental result: the unwashed labelled cells were not significantly slower in growth rate than thteir washed counterparts. This indicates that there is no influence from the fluorochrome-existed medium on cell growth. The protoplast releasing rate from the lebelled cells was over 98%. The cell-colony-formation rate of the FITC- and RITC-labelled protoplasts cultured in a liquid C₈IV medium was not different from that of the control (EtOH). Furthermore, the cell colonies developed from the RITC labelled protoplast at day 17 still emitted light pink color. The persisted existence of light pink color can serve as a visual marker for screening a true cell-fusion hybrid. |
