| 中文摘要 | 本研究在探討陰道鞭毛蟲釋放一種新的中性白血球激活因子(TV-NAF)對中性白血球趨化作用的影響。陰道鞭毛蟲(每毫升含10^7蟲體)在不含血清之漢氏緩衝溶液,於攝氏37度置放30分鐘後離心,製備為陰道鞭毛蟲上清液(STV)。應用多室(multi-chamber)趨化作用分析,發現陰道鞭毛蟲上清液對中性白血球之趨化指數可達112±15個細胞(400倍鏡檢),而對照之漢氏緩衝溶液只有11±4個細胞。若將陰道鞭毛蟲殺死則其上清液即失去對中性白血球之趨化活性,但此趨化活性不受加熱處理(高達攝氏100度)或過氯酸(4M)去蛋白質作用之影響,顯示陰道鞭毛蟲上清液所含之中性白血球激活因子本身不具蛋白質特性,由超濃縮法(ultrafiltration)可知此激活因子分子量低於3000道爾頓。因此,進一步以放射免疫分析法測定陰道鞭毛蟲廿酸物質(eicosanoids)之產生。結果可測得白球三烯達濃度500 pg/ml。本研究結果顯示陰道鞭毛蟲可自發性釋放出白血球三烯活化中性白血球,可能在陰道鞭毛蟲感染之致病機轉中扮演著某種角色。 |
| 英文摘要 | We have investigated effects of a novel neutrophil activating factor released by Trichomonas vaginalis (YV-NAF) on the neutrophil chemotaxis. TV-NAF was present in the supernatant from 107 T. vaginalis (STV) being in 1 ml of serum-free Hanks’ balanced salt solution (HBSS) at 37℃ for 30 min. Employing a multi-chamber chemotactic assay, as demonstrated by a chemotactic index showing migrated neutrophils were 112±15 (mean±SD, n=7) for STV and 11±4 for HBSS, per high power field (×400 magnification). There was no chemotactic activity in the preparation of STV from killed trichomonads. The fact that heating up to 100℃ or deproteinization by treatment with perchloric acid (4M) did not abolish its chemotactic activity of TV-NAF was associated with the fraction containing small molecules less than molecular weight 3000 daltons. Therefore, the possibility of eicosanoids production by trichomonads responsible for the neutrophil activation has been investigated. Leukotriene B4 (LTB4, 500 pg/ml) was found in the STV as demonstrated by radioimmunoassay. Production of LTB4 by trichomonads was time-dependent. These studies suggest that T. vaginalis may spontaneously release LTB4 to activate neutrophils and play a role in the pathogenesis of this trichomonad infection. |
