英文摘要 | The isoelectric focus electrophoresis was used to analyze seed proteins from 15 cultivars/lines of radish (Raphanus sativum). Seed proteins extracted by using 9M urea resulted the best IEF separation between PI 3.5 to 9.5. Seed proteins from radish were separated into eight sections: 0-2.2 cm, 2.2-2.8 cm, 2.8-3.4 cm, 3.4-4.0 cm, 4.0-4.9 cm, 4.9-5.7 cm, 5.7-6.9 cm, 6.9-10.0 cm, according to their migration distance from the origin on IEF gel. Only one scan of protein was observed in the first section, however, three scans, four scans, four scans, three scans, four scans, five scans, and three scans of proteins were detected in section 2 to 8, respectively. All the tested cultivars/lines were able to be distinguished by their IEF separated protein profiles. No significant relationships among protein profiles, intra leaf phenotypes, and root phenotypes of 15 cultivars/lines were detected. The same system was applied to analyze the seed proteins of garden pea (Pisum sativum and resulted seven mobile sections: 0-3.0 cm, 3.0-3.6 cm, 3.6-4.2 cm, 4.2-5.6 cm, 5.6-7.2 cm, 7.2-8.4 cm, and 8.4-10.0 cm, between PI 5 to PI 7. Five scans of proteins were observed in the first section followed with five, three, seven, five, five, and three scans of proteins in section 2 to 7, respectively. Similarly, all the tested cultivars could be identified by their protein profiles separated by IEF but no relationship between pod phenotypes and IFF protein profiles was observed. |
