| 中文摘要 | 人類膀胱癌細胞株(Yen-87, Zoa-88, J82, Shen-87 ),老鼠膀胱癌細胞株(MBT-2, M1660)及正常纖維細胞(FB-l,FB8490)被用於測試五種染料之光照殺死能力。結果發現血紫質HPD作用最強,其次為甲基藍,中等作用程度者為acridineorange,最弱者為eosin-B及維他命B2。細胞漿內乳酸去氫酵素活力之測定,體外雙層軟膠細胞簇落實驗皆顯示甲基藍或HPD染色後照光對多種癌細胞可破壞其酵素活力,軟膠內細胞簇落形成之能力。最適當之染色時間為30分,而最佳之照光時間則在60-90分之間,時間愈長,效果愈佳。染色照光之毒殺作用不僅止於膀胱癌細胞,對正常之纖維細胞也有相同作用。 |
| 英文摘要 | Human bladder cancer cell lines, J82, Yen-87, Shen-87 and Zoa-88, and murine bladder cancer celllines, MBT -2 and M1660, were used as the target cells for the dye-sensitized photoinactivation study in using different dyes. Normal fibroblast cells, FB-l and FB8490, were used as the control group. The cytoplasmic activity of lactic dehydrogenase and double layer soft agar clonogenic assay were measured and compared after in vitro photoinactivation between different dye reagents and cell lines. The dyes included hematoporphyrin derivative (HPD), methylene blue, acridine orange, eosin B and riboflavin. The cytotoxicity of photoinactivation was closely related to the duration of illumination. From both in vitro studies, it revealed the HPD had the highest cytotoxic activity against the bladder cancer cells then followed by methylene blue (strong activity), acridine orange (intermediate activity), eosin B and riboflavin (weak activity). More than 90 per cent of cytotoxicity could be obtained within 60 minutes of illumination after staining with HPD for 30 minutes. Normal fibroblast cells also demonstrated similar responses to the photoinactivation of various dyes as compared to that of cancer cells (Zoa-88, MBT-2 and M1660). These preliminary results suggest that HPD or methylene blue may be useful as an adjuvant photochemotherapy for superficial bladder cancer. |
