中文摘要 | 本研究之目的係運用酵素免疫法來評估去礦物化和非去礦物化之人類牙根表面對纖維粘蛋白之結合能力。我們取用牙周病患所拔牙齒之牙根,去除其牙骨質和牙髓組織,再將其切成薄片。這些牙根薄分成四組,第一組以飽和檸檬酸(PH值為1)處理十分鐘(CA10),第二組以飽和檸檬酸處理三十分鐘(CA30),第三組以12%eDTA加以?理三十分鐘(EDTA30),第四組不作任何處理(NONE),每組再分成兩小組:(1)用纖維粘蛋白處理過的(FN),(2)未南畢纖維粘蛋白處理過的(CONTROL),然後所有牙根薄片和1%bSA作用1小時,接著運用酵素免疫法評估纖維粘蛋白和牙根表面的結合量,結果顯示用檸檬酸處理十分鐘的牙根表面對纖維粘蛋白之結點能力是非去礦物化之牙根表面的兩倍,不過隨著處理時間的增長而結點能力有變弱的趨勢,另外用EDTA處理對牙根表面的纖維粘蛋白結合能力並沒有影響,由此可知用檸檬酸處理牙根表面可以促進纖維貼蛋白的附著。 |
英文摘要 | The aim of this study was to assess the binding ability of fibronectin (FN) to both undemineralized and demineralized human root surfaces by ELISA. Discs of dentine with the cementum and pulpal tissue removed were prepared from the roots of periodontitis-involved human teeth. The discs were divided into four groups: (1) treatment with saturated citric acid (CA, PH-1) for min (CA-10); (2) treatment with saturated CA for 30 min (CA-30); (3) treatment with EDTA (12% w/v) for 3- min (eDTA-30); (4) FN treatment (FN); (B) no FN-treatment (CONTROL). All discs were indubated for 1 h with 1 % BSA. The amount of fibronectin bound to the dentine surface was assessed by ELISA. The results revealed that the binding ability of FN in the CA-10 group was two-fold greater than that of the NONE group. But FN-binding ability decreased with the increased duration of CA-treatment. We found that the binding ability of FN on root discs was unaffected by EDTA treatment. Our results suggest that CA treatment of exposed roots may enhance the adhesion of FN. |
