中文摘要 | 本試驗目的在於瞭解低溫對豬胚存活之影響,特設計將豬胚冷却至15℃以下,並體外培養或移置,以觀察豬胚之存活情形。 本試驗使用7~8個月齡的新母豬,經由孕馬血清激性腺素(PMSG)和人類絨毛膜激性腺素(hCG)誘發超級排卵後配種、採胚。將獲得豬胚冷却至15和6℃ 2-18小時或給予冷凍至-35和-196℃,經冷却或冷凍的豬胚均使用37℃回溫和解凍,最後將豬胚行體外培養或直接給予移置,找出冷却或冷凍豬胚未能成功的瓶頸。 試驗結果如下: 1.誘發超級排卵所使用之PMSG和hCG,以PMSG 1750 I.U.和hCG 500~750 I.U.肌肉注射新母豬可獲得較穩定的效果。 2.15℃冷却桑椹期和囊胚期豬胚2個小時,經體外培養仍可存活24小時。冷却15℃ 2小時囊胚期形態正常的豬胚經移置,其成功率為50%(2/4),胚的存活率為15%(18/120)。 3.6℃冷却囊胚期和孵化囊胚期2個小時,經體外培養24小時,孵化囊胚期有較高的存活率50%。冷却6℃ 2小時孵化囊胚期形態正常豬胚經移置,其成功率為14.3%(1/7),胚的存活率為4.6%(7/151)。 4.擴張和早期孵化囊胚期豬胚冷凍至-35℃後解凍,經體外培養仍有60%的豬胚存活。但是冷凍至-196℃的豬胚則無存活。 由以上的結果顯示較後期的豬胚對冷似乎有較高的忍受性,所以如何選擇适合於冷凍的豬胚時期為其能否冷凍成功的先決條件。 |
英文摘要 | Pig embryos were extremely sensitive to low temperature, no pig embryo has been frozen (-196°C) alive successfully in so far. An attempt to preserve pig embryos below 15°C was conducted with some success. Those embryos were obtained from gilts of 7-8 months of age and were treated with hormone for superoverlation were mated artificially inseminated. Embryos were collected 5-7 days after hormone treatment by surgical operation. Morula to hatched blastocyst pig embryos were cooled to 15°C, 6°C for 2 hours or frozen to -35°C and -196°C, then incubated in 37°C, 5%CO₂ or transfer to a recipient gilt directly. The result were: 1. PMSG 17501, U and hCG 500-7501. U. for superovulation in gilts had a good result. 2. Pig embryos at the stage of morula and blastocyst after cooling to 15°C for 2 hours, its still could survive for 24 hours under incubation. If cooled embryos were transfer to recipient gilts, conception rate was 50% (2/4) and embryos survival rate was 15% (18/120). 3. Pig embryos at the stage of blastocyst and hatched blastocyst after cooling to 6°C 2 hours, hatched beastocyst had higher survival rate than blastecyst after 24 hours under incubation. If cooled embryos were transfer to recipient gilts, conception rate was 14.3% (1/7) and embryos survival rate was 4.6% (7/151). 4. Pig embryos at the stage of expand blastocyst and early hatched blastocyst after frozen to -35°C, 60% embryos still could survival for 24 hours under incubation, but no survive if frozen in -196°C. The experiment indicated pig embryos still tolerance to low temperature, but after cooling or frozen (35°C) and thawing, survival rate of late stage pig embryos were higher than early stage pig embryos. So, select suitable stage of embryos for cooling or frozen are importance, espicially of the survival of embryos after long term storage at -196°C in liquid nitrogen. |
