| 中文摘要 | 植物之原生質體可經誘導發生融合,融合反應不受種間的限制,融合細胞經培養後,仍具有分化全能性(totipotency),因此利用不同種間原生質體進行融合,培養與植株再生,將可育成體細胞雜種植物。本試驗即以野生種之菸草Nicotiana sylvestris與N. tomentosiformis之葉肉原生質體為材料,利用化學藥劑誘導融合,探討不同誘導條件對原生質體融合與融合產物生育之影響,藉以了解利用原生質體融合的方法,進行體細胞雜交之可行性。 由本試驗之結果顯示,原生質體在4×10⁵ pts/ml之密度下,添加10.5 mM氯化鈣做誘融劑,採用培養皿法,對原生質體的誘融效果最佳。此外,若同時於PEG溶液中再添加10%DMSO,亦可得較佳之融合效果。 經培養試驗發現,以含10.5 mM氯化鈣之50%PEG 6000溶液誘融之融合產物,在培養初期,細胞即發生質離或皺縮,導致死亡。而以含10.5 mM氯化鈣之25%PEG 6000溶液誘融,較有利於細胞分生。 |
| 英文摘要 | Plant protoplasts can be induced to undergo intra- and interspecific fusion. Fused protoplasts in culture are capable of dividing and forming cell colonies and even producing complete plants. This technique, named parasexual hybridization, is a new method to obtain hybrid plants without sexual crossing. This experiment was conducted to study the fusions of protoplasts from Nicotiana sylvestris and N. tomentosiformis. Suitable conditions of PEG (polyethylene glycol) - induced protoplast fusion was examined. It was found that high protoplast fusion frequency can be obtained when protoplast density is adjusted to 4×10⁵ pts/ml and treated with 50% (w/v) PEG 6000 solution supplemented with 10.5 mM CaCl₂.2H₂O and 10% DMSO. However, protplasts fused by treating with 25% (w/v) PEG 6000 solution supplemented with 10.5 mM CaCl₂.2H₂O showed betterviability in terms of division and colony formation during culture. |
