中文摘要 | 本實驗研究的目的是運用離子選擇性微電極與普通微電極,直接測量骨骼肌細胞膜電位和細胞內鈉離子活性濃度;以及細胞膜電位和細胞內鈉離子活性濃度相互間的關係。將大白鼠橫膈肌固定於灌流槽中,以Tyrode氏溶液灌流。於六隻大白鼠中,橫膈肌細胞膜電位為-86.1±4.5 mV,胞內鈉離子活性濃度為16.5±1.7 mM。當橫膈肌於低鈉溶液(20 mM)中 ,細胞膜電位由原來的-80.4±3.2 mV變為-83.4±7.5 mV,細胞內鈉離子活性則由原來的16.5±2.0 mM減少為11.9±3.0 mM (n=3,p<0.05);當橫膈肌於高鉀溶液(62 mM)中,細胞膜電位由-91.9±3.7 mV去極化為變為-59.0±5.2 mV (n=3,p<0.05),細胞內鈉離子則由原來的16.4±1.4 mM減少為10.9±0.6 mM (n=3,p<0.05)。實驗結果顯示細胞內鈉離子的活濃度隨著鈉離子在細胞膜上電氣化學位能差的降低而減少。 |
英文摘要 | The purpose of this study was to measure intracellular sodium ion activities in skeletal muscle and to explore the relationship between cell membrane potential and sodium ion activity. A piece of diaphragmatic muscle was fixed on the floor of perfusion chamber and circumfused with Tyrod solution. In six different preparation, the cell membrane potential was –86.2±4.5 mV, cytosolic sodium ion activity was 16.5±1.7mM in control condition. When the muscle was in condition of low [Na⁺]₀ (20mM), cell membrane potential changed from control – 80.4±3.2mV to – 83.4±7.5mV, cytosolic sodium ion activity decreased from 16.5±2.0mM to 11.9±3.0mM (n=3, p<0.05); when the muscle was in circumfusion of high [K⁺]₀ (62mM), cell membrane potential depolarized from control -91.9±3.0mV to -59.0±5.2mV, cytosolic sodium ion activity decreased from 16.4±1.4mM to 10.9±0.6mM (n=3, p<0.05). These results reveal that intracellular sodium ion activity parallels with decrement of the electrochemical gradient of sodium across the cell membrane. |
