中文摘要 | 利用活腦切片以及細胞内電生理記錄的技術,作者研究在大白鼠藍斑核神經細胞中,由地嗎啡所引起的高極化作用。我們使用兩種鉀離子通道的阻斷劑--氯化銫及蜂胺來確定地嗎啡所打開的鉀離子通道。此外,我們也以環腺甘酸的替代品--八溴環腺甘酸來灌流藍斑核神經細胞,以測試鴉片藥物引起藍斑核神經細胞高極化是透過環腺甘酸減少之作用的假說。實驗結果顯示,氯化銫可阻斷地嗎啡引起的藍斑核細胞高極化,而蜂胺則不能。因此我們認為在藍斑核細胞的細胞膜上被地嗎啡所打開的鉀離子通道為内向整流性鉀離子通道,而非SK型的依鈣性鉀離子通道。在八溴環腺甘酸的實驗上,即使在灌流八溴環腺甘酸的情況下,地嗎啡仍然可以使藍斑核神經細胞高極化。因此,我們認為環腺甘酸的減少在地嗎啡所引起的藍斑核細胞高極化的過程中並不佔有重要的角色。 |
英文摘要 | The dermorphin-induced hyperpolarization in rat locus coeruleus (LC) neurons was studied with techniques of brain slice and intracellular recording. Cesium chloride (CsCl) and apamin are capable of blocking the potassium channels of different types. These two kinds of potassium channel blockers were used to characterize the potassium channels opened by dermorphin. In addition, the LC neurons were peroused with 8-Br-cAMP to test the hypothesis that a decrease in intracellular cAMP mediated the opiate-induced hyperpolarization. The results showed that CsCl blocked the dermorphin-induced hyperpolarization, and apamin did not. This finding suggests that the inward-going rectification potassium channels, rather than the SK type calcium-dependent potassium channels are opened by dermorphon. Regarding the effects of 8-Br-cAMP, we found that even when the intracellular concentration of 8-Br-cAMP was increased, hyperpolarization still occurred, though to a lesser degree. Accordingly, a decrease in intracellular cAMP seems not to play an important role in the dermorphin-induced hyperpolarization of LC neurons. |
