中文摘要 | 由於人類胃癌研究進行緩慢,資料罄然,本研究室即由建立國人胃癌細胞株開始,進行系列之胃癌研究,期能對胃癌之防治,有所助益。本研究即以Köhler及Milstein的融合瘤技術,將自行建立的胃癌細胞株SC-M1,免疫BALB/C小白鼠,經多次的篩選(screening)及單株選擇(cloning)挑出二株融合瘤Ny-5與An-21,可產生抗體對抗胃癌組織細胞膜萃取物或胃癌細胞株抗原,但不能對抗纖維母細胞抗原。以酵素免疫分析法(enzyme-linked immunosorbent assay; ELISA),間接免疫螢光法(indirect immunofluore scence; IIF)測定此抗體與其他腫瘤細胞株之反應,並以過氧化酶-抗過氧化酶免疫複合體(peroxidase-anti-peroxidase)在冰凍切片觀察抗體之專一性及其在各種組織之分布情形。結果顯示此抗體與部分癌組織有交叉反應,並證實了抗體與腫瘤胚胎抗原(carcinoembryonic antigen; CEA)有結合之效應。此抗體應能提供日後有關腸胃道癌症之研究應用。 |
英文摘要 | Systematic investigations on human stomach cancer have been a goal for many researchers. However, there is lack of academic information on stomach cancer in Taiwan. We fortunately established a stomach cancer cell line SC-M1 from a patient in our laboratory recently, so as production of the monoclonal antibodies using SC-M1 cells was attempted. Two monoclonal antibodies were developed by the mouse-mouse hybridoma techniques. Their isotypes were IgG1, and the titre retained at as high as 10000. Additivity test indicated that those two antibodies probably recognized the same antigenic region. Indirect immunofluorescence and peroxidase-anti-peroxidase complex tests disclosed that the antigen recognized by the monoclonal antibody might be a common antigen. Further studies revealed that the antigen could be CEA-related, although there is no CEA secreted by SC-M1 cells. The antibodies were thus postulated having the metabolic pathway of CEA intracellularly. |
