中文摘要 | 著者等曾報告大鼠經ACTH長期處理後,對於腦下腺LH合成及釋出之影響,有性別間之差異(5)。本試驗之目的乃進而研究ACTH對於下視丘內激性腺素釋素(GnRH) 之影響。 選用正常及去腎上腺之雄鼠與雌鼠,每天皮下注射ACTH 0.2 mg,連續14天。於試驗期之末,取出各鼠之下視丘,應用放射線免疫法(RIA)測量其激性腺素釋素之含量。 具試驗結果,顯示對照組與處理組間,無論雄鼠或雌鼠,其下視丘內GnRH之含量,均無顯著之變化。由此證明ACTH對於大鼠生殖系統之影響,可能僅及於腦下腺階段,此亦?表示高量ACTH 或可減低腦下腺對於GnRH之感受性。此外,雄鼠或雌鼠經割除腎上腺後,如給與生理鹽水,其下視丘--腦下腺--性腺軸之功能,亦不被腎上腺割除之長期影響所改變。 |
英文摘要 | Sex difference in LH synthesis and release of pituitary gland response to chronic ACTH treatment was evidenced in rats by our previous report. The purpose of this study was to investigate whether the hypothalamic GnRH content was involved. Both male and female intact and adrenalectomized rats were used and injected subcutaneously with 0.2 mg of ACTH daily for 14 consecutive days. At the end of experimentation, the hypothalamus of rats were removed and the GnRH content was measured by radioimmunoassay. The results showed that there were no significant changes in hypothalamic GnRH contents between the control and treated groups in rats of both sexes. These findings suggest that the inhibitory effects of ACTH treatment on the reproductive system of rats only occur at the pituitary level. It also implies that excess ACTH amy be able to decrease the sensitivity of pituitary gland to GnRH. In addition, the function of the hypothalamo-pituitary-gonadal axis in rates of both sexes is little altered by chronically adrenalectomized treatment if the rats have been provided with physiological saline. |
