中文摘要 | 自綠豆(Phaseolus radiatus)下胚軸萃取的粗酵素液內含有IAA氧化酶的活性,IAA氧化酶的活性因加入輔因子Mn2+及2,4─雙氯酚(2,4-dichlorophenol)而大為增進。Mn2+及2,4─雙氯酚最適濃度分別為0.1 mM及0.05mM. pH值以5.6~6.0為最適宜。 在綠豆發芽期間,下胚軸IAA氧化酶的活性每天都在改變,但不與其長度成固定的比例;此時照光植物之酵素活性很低,因為此酵素液內含有大量的天然抑制劑,而黑暗培養之植物酵素的活性很高,內含之天然抑制劑較少,經透析後酵素活性不呈現靜止期。 當反應溶液內含有0.0006μmole核黃素或0.0001%過氧化氫時,對IAA氧化酶的活性有增進作用,似乎綠豆之IAA氧化酶的作用與過氧化酶有密切的關係。 |
英文摘要 | The erude enzyme extracts from the hypocotyls of Phaseolus radiatus (green bean) contained the IAA oxidase system and its activity was enh-anced by the addition of Mn2+ and 2,4-dichlorophenol (DCP). The opti-mal concentration of Mn2+ and DC P were 0.1 mM and 0.05 mM respectively. The pH optimum was 5.6~6.0. During various growth periods, the activity of IAA oxidase changed everyday, but it was not proportion to the length of the hypocotyl. The enzyme extracts from light- grown plants showed very low IAA oxidase activity, because the enzyme extracts contained an endogenolls inhibitor. The enzyme extracts from dark- grown plants contained very active IAA oxidas activity which had no lag phase. The results showed that in the presence of 0.0006μmole riboflavin or 0.0001 % H2O2 in the mixtures. the IAA oxidase activity was enhanced greatly. It seemed that the IAA oxidase of Phaseolus radiatus nayassociate with the peroxidase in performing IAA oxidation reaction. |
