中文摘要 | 本試驗以落花生栽培種與野生種種間雜交之子房柄為材料,探討在MS培養基添加植物生長調節劑、酪素水解物(casein hydrolysate, CH)及抗壞血酸(ascorbic acid)對促進子房柄形成莢果、胚發育及再生種間雜種植株的效果,期望建立種間雜交子房柄培養技術,作為克服落花生種間雜交不親和性之參考。 調查授粉後14天之母本(臺南11號)與種間雜交之子房柄培於添加150 mg/lCH及60 gl/sucrose的MS培養基120天得知,臺南11號之莢果形成率為50%,大多數種間雜交子房柄僅呈伸長反應及少數培植體發根。再添加1 mg/l NAA、0.05 mg/l BA及0.5 mg/l G□等植物生長調節劑,臺南11號之莢果形成率顯著降低,種間雜交子房柄均因誘導大量癒合組織而發育不良,僅有少數子房柄有膨大現象。 將經MS + 150 mg/l CH + 1 mg/l NAA+0.05 mg/l BA + 0.5 mg/l GA培養120天後仍有活性之種間雜交子房柄繼代培養於MS + 150 mg/l CH + 50 mg/l ascorbic acid培養基,有五個雜交組合之子房膨大且發育成莢果;再將 A. hypogaea × A. duranensis、A. hypogaea × A. pusilla及A. hypogaea × A. batizocoi三個種間雜交組合受粉後14天之子房柄培養於MS + 150 mg/l CH +50 mg/l ascorbic acid培養基亦均獲得莢果。種間雜交子房柄培養所得之雜交種子經無菌播種,A. hypogaea × A. cardensaii、A. hypogaea × A. batizocoi及A. hypogaea × A. pusilla三個組合均獲致再生植株。該等再生植株經染色體鑑定結果為2n=3x=30的三倍體雜種,生釀型式為類似野生種的分枝匍匐型。 |
英文摘要 | In order to establish the best protocol of peg tips culture to overcome cross-incompatibility between cultivated and wild species of Arachies, experiments were designed to enhanced pod-set and embryo development for obtaining interspecific hybrid plant. Fourteen-day-old peg of A. hypogaea (TN11) and hybrids crossed with nine wild species were cultured on MS medium with 60 g/L sucrose and 150 mg/L casein hydrolysate (CH) for 120 days, and 50% of pod formation were obtained in TN11 but only elongation and few rooting resulted the devoid of pod and swelling ovary in interspecific hybrid peg. Application of plant growth regulators (1 mg/L NAA, 0.05 mg/L BA and 0.5 mg/L G□) to the medium (MS + 60 mg/L sucrose + 150 mg/L CH) significantly decreased the rate of pod-set in TN11, and some of swelled ovule obtained in interspecific hybrids due to produce more callus and fewer roots. The viable peg tips of interspecific hybrid, cultured on the medium (MS + 150mg/l CH, 1mg/l NAA + 0.05mg/l BA + 0.5mg/l G□) for 120 days, transferred to the medium (MS + 150mg/l + 50mg/l ascorbic acid) and developed to pods for five crosses. The 14-day-old peg tips of A. hypogaea × A. duranensis, A. hypogaea × A. pusillaand A. hypogaea × A. batizocoi, cultured on the MS medium with 150 mg/l CH and 50mg/l ascorbic acid for 120 days, also developed to pods. The seeds derived from A. hypogaea × A. cardensaii, A. hypogaea × A. batizocoi and A. hypogaea × A. pusilla, germinated by aseptic seeding, and hybrid plants have been obtained. The results of cytological determination indicated that these hybrids with chromosome number 2n=2x=30 were triploid, and displayed the branch procumbent type tended to wild species. |
