| 中文摘要 | 人類多瘤性病毒,BK病毒,主要殼體蛋白VP1可在酵母菌系統(Saccharomyces cerevisiae INVSC1)成功的表達,且自我組裝成類病毒顆粒(virus-like particles; VLPs)。由穿透式電子顯微鏡觀察,此類病毒顆粒類似真的病毒顆粒(virion)具有血球凝集(hemagglutination assay;HA)的活性,能與人類O型紅血球產生凝集。應用此類病毒顆粒感染猴子之腎臟細胞(Vero cells),發現其感染後可很快進入細胞,且成功包裝螢光基因ODN-FAM進入Vero cells細胞中。此結果顯示,將來可應用此BKV VP1 VLP當作基因治療之載體,攜帶藥物治療多瘤性病毒相關的腎病變(polyomavirus-associated nephropathy)。 |
| 英文摘要 | Objective: The major capsid protein, VP1, of the human polyomavirus BK virus was expressed in yeast (Saccharomyces cerevisiae INVSC1). VP1 protein expressed in yeast was able to assemble into virus-like particles (VLPs). The VP1-VLPs was similar to BK virion in morphology as demonstrated by transmission electromicroscopy. VP1- VLPs could also cause haemagglutination of human O-type red blood cells. In the functional studies, the Vero cells of monkey kidney cells was infected with BKV VP1 VLPs. The result demonstrated that VP1-VLPs was able to enter the Vero cells. The BKV VP1 VLPs was also able to pack and deliver the ODN-FAM into the cell of Vero cells. These evidences showed that BKV VP1 VLPs might become a promising tool for the gene therapy and drug-diliver to treat the pathogenesis of PVN (polyomavirus associated nephropathy). |
