| 中文摘要 | 對於細菌的菌種,脈衝式電泳法(Pulsed-field gel electrophoresis,PFGE)是一種具有高度鑑別亞型的方法。為了增加脈衝式電泳法的鑑別力,我們評估AscI,NotI,SfiI,SmaI,ApaI and SgrAI六種不同的限制酶。在實驗中總共收集33株鮑氏不動桿菌(A.baumannii),這些菌株的取得是從2006年1月到10月在大林慈濟醫院住院的病人之臨床檢體,使用VITEK® 2 system做鮑氏不動桿菌菌株的鑑定。使用NotI,SfiI,SgrAI和Asc I等限制酶進行PFGE types的分析結果顯示具有高度的鑑別力(D≧0.99),在這四種酵素中以AscI產生最佳的可顯現之基因片段同時也顯示AscI也是最高的鑑別力之一;而ApaI和SmaI則顯示有近似的鑑別力。這結果顯示Asc I的鑑別力優於ApaI和SmaI。雖然所有的限制酶(NotI,SfiI,SmaI,ApaI及SgrAI)進行A.baumannii菌株PFGE types的分析皆可獲得令人滿意的結果,我們建議在脈衝式電泳法的分析步驟中也可以使用AscI限制酶來區別鮑氏不動桿菌之菌種。 |
| 英文摘要 | Background: Pulsed-field gel electrophoresis (PFGE) is a highly discriminatory subtyping method for bacterial species. To increase discriminatory index of this method, six restriction enzymes with Apa I, Sma I, Not I, Sfi I, SgrA I, and Asc I were evaluated. Thirty-three A. baumannii isolates were obtained at Buddhist Dalin Tzu Chi General Hospital from January to October 2006 and their identities confirmed using the VITEK® 2 system. The results of PFGE analysis showed that Not I, Sfi I, SgrA I, and Asc I exhibited high discriminatory index (D≧ 0.99). The Asc I generated the optimal visualization of genomic fragments and PFGE analysis with Asc I also exhibited the one of highest discriminatory index. The discriminatory index of PFGE analysis with Apa I and Sma I were similar. The results reveal that the discriminatory index of Asc I are superior to Apa I and Sma I. Although all enzymes produced satisfactory results, we also suggest Asc I may be used in a PFGE protocol for distinguishing A. baumannii isolates. |
