中文摘要 | 鵝是具有光週期依賴性且可預測的季節性繁殖物種,由於鵝對光變化的強烈反應使其繁殖性能較其他家禽差, 這些繁殖性能的變化主要受到下視丘-腦垂體-性腺軸的調控。本研究之目的為探討白羅曼公鵝在不同繁殖期下視丘蛋 白質之差異表現。試驗使用 9 隻三歲齡白羅曼公鵝,飼養在自然光照週期下,分別於非繁殖期、性腺恢復期與繁殖期 犧牲採集下視丘供蛋白質體分析。下視丘樣品以二維差異膠體電泳(two-dimensional difference gel electrophoresis, 2D-DIGE)進行蛋白質體的分析,差異表現蛋白質(differentially expressed proteins, DEPs)以質譜法進行身分鑑定及 以 gene ontology (GO)與 KEGG pathway 資料庫進行基因註解與生物功能路徑分析。試驗結果顯示,在所有定量的 1,537 個蛋白質點中,不同繁殖期公鵝下視丘有 147 個 DEPs,經質譜分析成功鑑定到 99 個蛋白質身分,並分屬於 64 個不同蛋白質;GO 基因註解顯示這些 DEPs 大多與蛋白質及離子的結合等分子功能有關,並參與細胞過程、代謝過 程及生物性調節等生物過程;功能性路徑分析顯示這些 DEPs 與檸檬酸循環 ( DLD、DLST、ACO2、SDHA 及 MDH2)、 糖解作用/糖質新生( DLD、ENO2、ALDOC 及 LDHA )、間隙連接( TUBA8、TUBB2B 及 TUBA1C)以及氧化磷酸化 ( UQCRC1、UQCRC2、ATP5B、SDHA 及 ATP6V1B2)有關。由本研究結果可知,繁殖期會影響白羅曼公鵝下視丘蛋 白質的表現,不同繁殖間之 DEPs 可能影響到生殖相關上游內泌素的調節。 |
英文摘要 | Domestic geese are photoperiod-dependent species with predictable seasonal breeding behavior; the changes in reproduction are mainly regulated and controlled by the hypothalamic-pituitary-gonadal axis. In this study, the differential expression of proteins in the hypothalamus of White Roman ganders at different reproductive stages was investigated. Nine ganders (average age of 3 years) were reared under natural photoperiod, and the hypothalamus of three ganders was collected at non-breeding, sexual reactivation, and breeding stages. Tissues were subjected to proteomic analysis by two-dimensional difference gel electrophoresis. The differentially expressed proteins (DEPs) were identified by mass spectrometry and subjected to gene annotation through the Gene Ontology (GO) database and functional pathway analysis employing the Kyoto Encyclopedia of Genes and Genomes pathway database. Out of 1,537 quantified protein spots, there were 147 DEPs in the hypothalamus of ganders at different breeding stages. A total of 99 DEPs were identified, referring to 64 unique proteins. Most of the DEPs identified through GO are associated with the molecular functions of protein and ion binding and are involved in cellular processes, metabolic processes, and biological regulation. The functional pathway analysis revealed that these DEPs are related to the tricarboxylic acid cycle ( ACO2, DLD, DLST, IDH3A, MDH2, and SDHA), glycolysis/gluconeogenesis ( DLD andENO2), gap junction ( TUBA1C, TUBA8, and TUBB2B), and oxidative phosphorylation ( ATP5B, ATP6V1B2, SDHA, and UQCRC2) These data suggest that breeding stages affect the hypothalamic protein expression in White Roman ganders. The DEPs among different breeding stages may impact on the efficiency of neuron transmission and the regulation of upstream reproductive hormones. |
