中文摘要 | 類蕈孢素胺基酸(mycosporine-like amino acids,MAAs)為一種海洋生物胞內物質,具有吸收UV光的能力,以保護海洋生物避免受UV光傷害。本研究將評估4種台灣本土海藻(海木耳、菩提藻、紫菜、和礁膜)之MAAs總量,並評估3種不同MAAs萃取方法(水浴法、超音波輔助水浴法、和淋洗法),及其最佳萃取參數,包括不同萃取溶劑(甲醇、乙醇)、溶劑比例(0、20、40、100%)、和液固比(10、20、30、60和90)。本研究可成功從不同海藻(海木耳、菩提藻、紫菜、礁膜)中萃取MAAs,且萃出液之吸收光譜波長範圍約280-400nm。就不同萃取方法方面,淋洗法可較節省時間、人力及電力成本,且萃出效率與水浴法和超音波水浴法相似。於不同萃取條件方面,本研究發現以20%甲醇溶液搭配60倍液固比之Porphyra-334萃出總量最佳,最高可達5.98mg/g,且前3次之累積溶出比例即可達87%以上,前4次即可達90%以上。 |
英文摘要 | Mycosporine-like amino acids (MAAs) is a family of intracellular ultraviolet (UV)-absorbing compounds which protect aquatic organisms against solar UV radiation. This study aims to understand the total MAAs contents in four Taiwanese seaweeds (Porphyra, Grateloapia, Sarcodia, and Ulva), and evaluated three extraction methods (water bath, sonication bath and water flush), and their best extraction conditions including different solvents (methanol and ethanol), solvent ratios (0, 20, 40, and 100%) and liquid-to-solid ratios (L/S: 10, 20, 30, 60 and 90 times). Absorbance of various leachates extracted from the four Taiwanese seaweeds, ranged from 280-400 nm. With regard to the extraction methods for extracting MAAs from four seaweeds, the water flush was the best one for saving time, manpower, and electric power. With regard to the extraction conditions, flush method with 20% methanol and L/S of 60 is best for Porphyra-334 extraction (up to 5.98 mg/g). And, 87% of accumulated Porphyra-334 ratio was leached by 3 extracted times. More than 90% could be achieved by four extracted times. |
