中文摘要 | 本研究選殖東方果實蠅 (Bactrocera dorsalis) 血管收縮素轉化酶 (angiotensin-converting enzyme, ACE) cDNA (BdACE) (GenBank accession no. MH795420) 全長序列 2081 bp,其中 包含 1848 bp 之開放讀架,轉譯一個 615 個胺基酸之蛋白。BdACE 胺基酸序列包含與鋅離子接 合使酵素具有活性之胺基酸:甘胺酸 (glycine)、精胺酸 (arginine) 及谷胺酸 (glutamine),分別 位於胺基酸序列序號 374、378 及 399 的位置。利用即時定量聚合酶連鎖反應測試 BdACE 於各發 育時期表現量,結果顯示 BdACE 於幼蟲時期表現量從一齡幼蟲至三齡末期呈上升趨勢,且在三齡 末期為顯著最高。於蛹期階段,表現量在第一日齡蛹為顯著最高,其餘日齡則未有顯著差異。於成 蟲時期,雄蟲 BdACE 於脂肪體之表現量,在羽化後第零日齡表現量為顯著最高,於第一日齡至第 十日齡則無顯著差異。雌蟲 BdACE 於脂肪體表現量,在第零日齡到第十日齡間皆無顯著差異。上 述結果顯示,BdACE 在發育轉換時期表現量上升,推論和發育及變態有所關連。進一步以反轉錄 聚合酶連鎖反應偵測 BdACE 於成蟲各組織之表現,結果顯示 BdACE 表現於脂肪體及腦,推測參 與修飾脂肪體蛋白及神經胜肽。 |
英文摘要 | In this study, an angiotensin-converting enzyme cDNA of the Bactrocera dorsalis (BdACE) was cloned. The full length sequence of BdACE was 2,081 bp which included a 1,848-bp open reading frame that translated into a 615-amino acid protein. Three amino acids, namely glycine375, arginine379, and glutamate400, are responsible for binding to Zn ion to activate angiotensin-converting enzyme (ACE). Real-time quantitative polymerase chain reaction (PCR) was conducted to detect relative mRNA expression of BdACE in different developmental stages. In the larval stage, the relative mRNA expression of BdACE increased with age; especially in the late third instar, the expression was significantly high. In the pupal stage, the BdACE relative transcriptional profile of BdACE was as follows: 1-day-old pupae had significantly high level of expression, and no significant difference was noted in the expression among 0- and 2- to 8-day-old pupae. In male fat body, the relative transcriptional expression of BdACE after eclosion was significantly high, but no significant difference was observed among 1- to 10-day-old males. In female fat body, no significant difference was noted among 0- to 10-day-old females. The developmental transcriptional profiles of BdACE suggested that BdACE may be involved in development and metamorphosis because high expression levels were observed in the transition period. To detect the transcriptional expression of BdACE in different tissues, reverse transcriptionPCR was used. The results revealed that BdACE was expressed in fat body and brain. This suggested that BdACE palyed a possible role in processing of fat body proteins and neuropeptides. |
