中文摘要 | 本實驗目的欲探討餵食綠原酸咖啡對糖尿病大鼠傷口癒合之促進效應。SD雄性大鼠以Streptozotocin(STZ)誘導高血糖後隨機分成5組:糖尿病對照組(餵予RO水)、低、中、高綠原酸咖啡組(分別餵予含CGA 20、50、70 mg/kg BW之咖啡量)、純綠原酸組(餵予CGA 70 mg/kg BW);第6組為沒有糖尿病的大鼠作為正常對照組(餵予RO水)。所有大鼠於背部切除皮膚產生傷口後,即進行管餵並觀察傷口癒合情形。結果指出,綠原酸咖啡或綠原酸餵食對於糖尿病鼠的血糖、飲水量、排尿量及肝功能酵素皆無顯著影響。糖尿病對照組於第8天的傷口癒合面積百分率(約18%)有低於正常對照組(48%)之趨勢;經餵予咖啡或綠原酸的糖尿病鼠傷口癒合面積百分率約為37%,有較糖尿病對照組為高的趨勢。取各組第14天的癒合皮膚經組織切片染色後觀察,糖尿病鼠餵予咖啡或綠原酸後,血管密度有較糖尿病對照組少的趨勢;高綠原酸咖啡與綠原酸組則有較低的發炎細胞密度。本實驗指出,綠原酸咖啡或是綠原酸具有促進傷口癒合之潛力。 |
英文摘要 | The aim of this study was to evaluate the effect of chlorogenic acid (CGA)-rich coffee on wound healing in diabetic rats. Materials and methods: Hyperglycemia was induced with a streptozotocin (STZ) injection in male SD rats. Diabetic rats were divided into the following five groups: (1) DM-C, only fed water; (2) DM-CF (L), fed coffee with a low dose of CGA of 20 mg/kg body weight (BW); (3) DM-CF (M), fed coffee with a medium dose of CGA at 50 mg/kg BW; (4) DM-CF (H), fed coffee with a high dose of CGA at 70 mg/kg BW; and (5) DM-CGA, fed CGA only at 70 mg/kg BW. The 6^(th) group was normal rats (NC group) only fed water. The healing of a dorsal skin wound was observed every other day for 20 days. The wound skin on the 14^(th) day was excised for a histological analysis. Results: There were no significant differences in plasma glucose levels, volume of water intake and urine output, or the plasma enzyme activity for liver function among the five diabetic groups. The wound healing percentage of the DM-C group (18%) on the 8^(th) day was lower than that of the NC group (48%). Diabetic rats fed CGA-rich coffee or CGA showed an average percentage of wound healing of 37%. The histological analysis of the wound skin on the 14^(th) day showed fewer vessels and a lower inflammatory cell density in the high CGA-rich coffee and CGA groups. Conclusion: CGA or CGA-rich coffee consumption might have potent wound-healing properties in diabetic rats. |
