中文摘要 | 單核苷酸多態性(single nucleotide polymorphisms, SNPs)可以透過候選基 因方法用於遺傳標識輔助選拔,從而改進豬隻繁殖性狀。選擇Ataxia-telangiectasia mutated 基因(ATM gene)作為候選基因,因為它在豬隻桑椹胚至囊胚期之間具 有差異性表現(量),因此其可能在胚胎發育中扮演重要的角色。之前研究報告 顯示,在藍瑞斯(Landrace)母豬位於 ATM 基因 5' 側翼區域具有約 13 個SNPs。 本試驗研究利用基因定序及高通量的 12-pex GenomeLab SNPstream 基因型分析 系統,針對 456 頭藍瑞斯母豬進行 SNPs 基因型分析,並且將分析所得 ATM 基因 SNPs 基因型與試驗母豬的窩仔數及平均仔豬重等資料進行關聯性分析。試驗結果 顯示,在藍瑞斯母豬其 ATM 基因 SNPs 基因型 -A to J、-K 和 -M 與總出生仔豬數; SNPs 基因型 -A to J、-K 和 -L 與出生活仔數及 SNPs 基因型 -A to J 與平均出生仔 豬重間,皆具有顯著性的差異性(P < 0.05)。此外,具有調節性的 SNPs(ATM-B, A-105G)位置,可能被包含在一些潛在的轉錄因子結合位點(如:ZBPF 和 KLF),可能的功能為改變基因調控。此試驗研究結果顯示,ATM 基因的 SNPs 未 來可能可以作為臺灣藍瑞斯母豬繁殖性狀遺傳育種選拔的有效遺傳標識。 |
英文摘要 | Single nucleotide polymorphisms (SNPs) can be used for a marker-assisted selection through the candidate gene approach to improve the reproductive traits in pigs. The ataxia-telangiectasia mutated (ATM) gene was chosen as a candidate gene because it was differentially expressed between the morula and blastocyst stages and may play important roles in embryonic development. All the 13 SNPs located in the 5’-flanking region of the ATM gene were identified in Landrace sows. We analyzed the SNP genotypes for 456 sows by using sequencing and the GenomeLab SNPstream 12-plex high-throughput genotyping system. The SNPs of the ATM gene were tested for their association with recorded litter size and average birth weight. The results revealed that the SNP genotypes of the ATM gene were significantly associated with total number of piglets born (P < 0.05) for -A to J, -K, -M; number born alive for -A to J, -K, -L (P < 0.05) and average birth weight (P < 0.05) for A to J in Landrace sows. In addition, the regulatory SNP (ATM-B, A-105G) was involved in some potential transcription factor binding sites (ZBPF and KLF) which may functionally alter gene regulation. Our research illustrated that the SNPs of the ATM gene may be useful markers in the selection for improvement of reproductive traits in Landrace sows in Taiwan. |
