中文摘要 | 顆粒單核球群落刺激生長因子 (Granulocyte-macrophage colony-stimulating factor, GM-CSF) 藉CD34 +沿著不同的樹突細胞非依賴路徑加速催熟造血幹細胞,CD1a 和 CD14的族群數量倍增而CD14 + and CD1a - 的族群逐降消失,GM-CSF 對免疫功能的影 響如下:一、細胞激素調節;二、T細胞極化;三、平衡發炎介質,促成管制T細胞 的分化和成熟;四、誘導耐受樹突細胞;五、增加 GM-CSF 自體免疫容忍和減少異 體移植的排斥。GM-CSF 調理T細胞反應和藉著增生骨髓前驅細胞,影響移植排斥作 用的發生和嚴重度,且改變自體移植的過程。源自骨髓幹細胞的顆粒細胞藉由 GMCSF 的依賴機制,被腫瘤來源的啟動因子,藉 STAT3路徑把骨髓顆粒/吞噬細胞,脫 離常規地活化。癌症病人的兩組骨髓來源的幹細胞亞群有單核 CD14細胞和顆粒 CD15 細胞,徵招的骨髓源幹細胞和保持在腫瘤內的癌幹細胞,被誘導出細胞毒性風暴消 滅,就是藉 GM-CSF 的細胞毒殺能力作為癌症免疫療法之一。 |
英文摘要 | GM-CSF through CD34 + accelerate the ripening of hematopoietic progenitors differentiate along with various independent dendrite cell pathway in response to GMCSF. CD1a and CD14 double positive population, with progressive disappearance of CD14 + and CD1a -. Effects of GM-CSF on immune functions : (1) induce modulation of cytokines; (2) T cell polarization; (3) balance between inflammatory mediators, favor Treg differ and mobile; (4) induce tolerance DCs; (5) increase GM-CSF inhibit autoimmune and allergenic T cell response. GM-CSF modulate T cell react and effect incidence and severity of acute and chronic GVHD by expanding myeloid precursors, and alter allograft composition. Granulocyte MDSC develop via GM-CSF dependent mechanism. Aberrant myelopoiesis from bone marrow progenitors (i.e., granulocyte-macrophage /GMP) is initiated by TDF, through STAT3. Two main subsets of MDSC identified in cancer patient, a monocyte CD14 and a granulocyte CD15. MDSC recruit and maintain in tumor, strategy could be developed to reuse immune tolerance to tumor. |
