| 中文摘要 | 本計畫主旨為建立一系統化研究平台,包括標的藥用植物之基原鑑定、化學指紋圖譜分析及指標成分分析,並以具體細胞學 /分子生物學的生物活性測試平台及動物實驗模式,以確定台灣本土常用之藥用植物之物種、化學指紋圖譜、活性指標成分並利用實證醫學驗證其活性,以提升本土中草藥之應用價值。在台灣本土常用藥用植物基原鑑定研究上,我們完成狗肝菜之品種確認、組織切片鏡檢觀察及核醣體核酸基因 (rDNA)序列分析之分子基原鑑定檢測。在體外抗發炎及免疫調節活性測試上,我們以評估樹突狀細胞細胞激素調控做為初步體外篩選平台。 研究結果顯示,薑黃酒精萃取物可有效抑制細胞激素 IL-6及 IL-12之表現。大花咸豐草及狗肝菜酒精萃取物對於細胞激素 IL-6有部分抑制活性。另一方面,月桃、大花咸豐草及狗肝菜酒精萃取物對於細胞激素 IL-12皆具有有顯著之抑制活性,尤其以大花咸豐草酒精萃取物在高濃度 (50 g/ml)或是低濃度 (6.25 g/ml)皆具有顯著的抑制效果。在原位乳癌動物模型之抗腫瘤活性測試,薑黃酒精萃取物對於原位乳癌細胞生長有顯著的抑制活性;在腫瘤轉移結果則顯示,薑黃、大花咸豐草酒精萃取物與實驗對照組相比,皆具有抑制乳癌細胞轉移的效果。在乳癌切除動物模型中的抗腫瘤轉移活性上,結果再次驗証薑黃及大花咸豐草酒精萃取物可降低腫瘤轉移率及顯著提高老鼠存活率。由目前我們所得到的結果使我們相信此計畫所建立之研究篩選平台將有助於未來中草藥從種植、採收、品管到活性的垂直整合系統,產生一套能提供功效好且品質穩定之中草藥的標準化栽種生產程序(Standard Operating Procedure),進而促使台灣本土中草藥能逐步具體地科學化,以確保民眾使用之安全,並有助於本主中草藥之發展。 |
| 英文摘要 | The objective of this study is aimed to build a biotechnology platform for verification of the authenticity of germplasms and bio-activities/efficacy of Taiwan specialty medicinal herbs, by using a combinational approach of morphological/anatomical analyses, followed by molecular maker (eg., DNA sequence) authentication, and specific bio-activity assays. High Performance Liquid Chromatography (HPLC) and LC/MS profiling of secondary metabolites in test plants also is analyzed to establish the relationship among their specific phytochemical marker components, “the efficacy” and bio-activity of test herbal cultivars/species and the specific aboriginal vs “adopted” cultural environments. Our preliminary results showed and confirmed the morphological and histological characteristical of Dicliptera chinensis plants in Taiwan. Analysis of ribosomal DNA sequence of D. chinensis revealed significant differences between this plant and other DNA sequences of related plant species, based on the NCBI GenBank database. A number of the ethanol and ethyl acetate extracts of test Taiwan specialty medicinal herbs exhibited an inhibitory activity on LPS-induced expression of IL-6 and IL-12 in mouse bone marrow derived dendritic cells (BMDCs). In addition, these herbal extracts were categorized into several functional sub-groups based on their capacities to regulate cytokine expression. In an orthotopic breast tumor mouse model, the ethanol extract of Curcuma longa effectively suppressed primary tumor growth. The ethanol extracts of both Curcuma longa and Bidens pilosa plants suppressed mammary tumor metastasis, and the results were further confirmed by a mammary breast tumor resection animal model. Together, we believe that the systematic platform that we developed in the present study has provided us with a useful approach for future biotechnology development of traditional herbal medicines in Taiwan. |
