中文摘要 | 本文依據衛生署健康食品安全性評估規範,進行對大鼠(SD品系)之口服急毒性試驗,探討穿龍薯蕷(Dioscorea nipponica)對哺乳類動物之安全性。穿龍薯蕷(T101-144)為淡棕色粉末,試驗純度視為100%。試驗時穿龍薯蕷以逆滲透水最高可配製成溶液濃度為0.085g/ml,每隻大鼠灌食體積量為20ml/kg body weight,當日依體重經口強迫灌食大鼠3次,每次間隔4小時,最終投予記量總計為5g/kg body weight,投予後連續觀察14天。結果顯示,以穿龍薯蕷口服投予大鼠後,全部鼠隻無中毒症狀或死亡,顯示對大鼠之口服急毒性LD50值為大於5g/kg body weight。穿龍薯蕷處理組雄、雌鼠第0、7及14天時體重及增重並無影響。試驗結束後,檢查白血球及紅血球總數、血紅素、血球容積比、平均紅血球體積、平均血紅素與平均血紅素度、及白血球分類等血液值部分與對照組相比雖有統計學上顯著性增加或減少(P<0.05),但皆介於正常生理值內,並無與試驗物質毒性有關之影響。肝腎血清酵素值天門冬酸轉氨酶(AST)、氨基丙酸轉氨酶(ALT)、尿素氮(BUN)及肌氨酸(creatinine)等項目亦無與試驗物質有關之影響。體內臟器重量包括腦、心、肝臟、腎臟、脾臟、胸線、腎上腺、睪丸及卵巢及等重量及百分比與對照組比較無明顯差異。綜合以上試驗結果,穿龍薯蕷對大鼠之口服急毒性LD50值為大於5g/kg body weight。 |
英文摘要 | According to the Health food security assessment norms, Department of Health, an acute oral toxicity test in rats (SD strain) was performed in this study to investigate the safety of Dioscorea nipponica on the mammals. The powder color of D. nipponica (T101-144) was light brown, and considered to be 100% purity. A powder solution concentration of 0.085 g / ml can be formulated using RO water. According to the body weight, forced feeding volume capacity of 20 ml / kg body weight by mouth was given to each rat, three times in a day at intervals of four hours. The total dosing estimated about 5 g / kg body weight, and a continuous observation last for 14 days. The results showed that after oral administration, all rats had no poisoning or death symptoms, and indicated the rat oral acute toxicity LD50 value was greater than 5 g / kg body weight. After 0, 7 and 14 days, weight and weight gain of male and female rates treated with D. nipponica solution (T101-144) were not affected. After the test, although statistically significant increase or decrease (p <0.05) in part with the control group were found when compared to the following blood values, including total number of white blood cells and red blood cells, hemoglobin, hematocrit, mean corpuscular volume, mean hemoglobin average hemoglobin concentration and white blood cell classification, but these blood values were in the range of the normal physiological values, there was no toxicity on test substance.The values of liver and kidney serum enzymes, including aspartate aminotransferase (AST) , alanine aminotransferase (ALT) , blood urea nitrogen (BUN) and creatinine (creatinine) , showed no related effects on test substance. The weight of the body organs, including weight and percentage of brain, heart, liver, kidney, spleen, thymus, adrenal gland, testes and ovaries, when compared to the control group indicated no significant difference. Based on the above test results, rat oral acute toxicity LD50 value of D. nipponica was greater than 5 g / kg body weight. |
